A widely expressed free immunoglobulin ? chain with a unique V?4-1/J?3 pattern promotes colon cancer invasion and metastasis by activating the integrin?1/FAK pathway

Qianqian Wang et al.

Summary: In this study, a free Ig kappa light chain V kappa 4-1/J kappa 3-FLC, normally expressed in B lineage cells, was found to be widely expressed in non-B lineages and overexpressed in cancer cells, promoting cancer cell proliferation, migration, and metastasis by binding to integrin beta 1 and activating FAK and Src pathways. Specific antibodies against V kappa 4-1/J kappa 3-FLC variable region significantly inhibited colon cancer tumor growth.

Glomerular deposition of galactose-deficient IgA1-containing immune complexes via glomerular endothelial cell injuries

Yuko Makita et al.

Summary: Galactose-deficient immunoglobulin A1 (Gd-IgA1) plays a crucial role in the development of IgA nephropathy (IgAN). The study found that Gd-IgA1-containing immune complexes (ICs) have a high affinity for glomerular endothelial cells, leading to dysfunction of the glomerular filtration barrier mediated by glycocalyx loss. This enhances the permeability of antibodies in the mesangial region and induces inflammatory responses in the pathogenesis of IgAN.

Downregulation of PPAR alpha mediates FABP1 expression, contributing to IgA nephropathy by stimulating ferroptosis in human mesangial cells

Jingkui Wu et al.

Summary: IgA nephropathy (IgAN) is the most common primary glomerulonephritis and a major cause of end-stage renal disease. The downregulation of PPAR alpha pathway affects FABP1 and GPX4 levels, leading to ferroptosis in human mesangial cells (HMCs) and contributing to the pathogenesis of IgAN.

Activated mesangial cells acquire the function of antigen presentation

Yinghua Zhao et al.

Summary: Mesangial cells (MCs) are resident cells of the kidneys that play a crucial role in maintaining glomerular function. They can be affected by changes in the glomerular environment, leading to functional transformation and involvement in the pathogenesis of various kidney diseases. Research suggests that activated MCs can act as antigen-presenting cells (APCs) in renal diseases.

Role of Human Mesangial-Tubular Crosstalk in Secretory IgA-Induced IgA Nephropathy

Junjun Zhang et al.

Summary: This study found that approximately one-third of IgAN patients had SIgA deposition in the mesangium, but SIgA was rarely detected in the tubulointerstitium. The binding rate of SIgA to human renal mesangial cells was significantly higher than to human proximal tubule epithelial cells. Inflammatory factors released from the mesangium via mesangial-tubular crosstalk may mediate tubulointerstitial damage in IgAN.
