Validation of carbon isotopologue distribution measurements by GC-MS and application to 13C-metabolic flux analysis of the tricarboxylic acid cycle in Brassica napus leaves

Summary: The estimation of metabolic fluxes in photosynthetic organisms is a significant challenge, especially with the development of C-13-Metabolic Flux Analysis. To accurately measure Carbon Isotopologue Distribution and minimize uncertainties for the calculated fluxes, the accuracy of GC-MS at the isotopologue level needs to be evaluated. This study developed a workflow to validate GC-MS measurements of Carbon Isotopologue Distributions in plant metabolites by using standardized extracts and investigated the light/dark regulation of the TCA cycle in Brassica napus leaf discs. The findings showed the cyclic operation of the TCA cycle under both light and dark conditions, with interesting contributions of different pathways and reassimilation of carbon derived from mitochondrial enzymes.

Integrated flux and pool size analysis in plant central metabolism reveals unique roles of glycine and serine during photorespiration

Summary: Photorespiration is an essential process in plant carbon and nitrogen metabolism that recycles inhibitory intermediates and provides carbon and nitrogen for metabolism. This study used isotopically non-stationary metabolic flux analysis to measure amino acid export rates and found that about 23-41% of photorespiratory carbon is exported as serine. The accumulation and relaxation of glycine pools were also found to constrain photosynthetic acclimation during photorespiratory transients.

Metabolic flux analysis of the non-transitory starch tradeoff for lipid production in mature tobacco leaves

Summary: The metabolic plasticity of tobacco leaves was demonstrated by generating transgenic plants with high lipid accumulation. Research showed that foliar lipids increased gradually with plant development, while starch content decreased, indicating a metabolic carbon tradeoff between starch and lipids. Additionally, the contribution of NADP-malic enzyme to plastidic pyruvate production for lipid synthesis was highlighted as a key factor in the successful leaf lipid engineering efforts in tobacco.

Exact mass GC-MS analysis: Protocol, database, advantages and application to plant metabolic profiling

Summary: Plant metabolomics is widely used in plant physiology to study metabolic responses to environmental parameters. Derivatization followed by GC-MS metabolic profiling is a major technique for quantifying primary carbon, sulphur, and nitrogen metabolites. However, routine exact mass GC-MS analyses are not common due to the lack of dedicated databases. In this study, a curated database for exact mass metabolomics focused on primary metabolism was presented. The advantages of exact mass analyses were demonstrated, including resolving isotopic patterns, localizing S-containing metabolites, and avoiding identification errors. A practical example using Arabidopsis leaves showcased how exact mass GC-MS analysis can be used for plant samples and identify metabolic profiles.
