Biological Effects of Indole-3-Propionic Acid, a Gut Microbiota-Derived Metabolite, and Its Precursor Tryptophan in Mammals' Health and Disease

Piotr Konopelski et al.

Summary: This review investigates the biological actions of indole-3-propionic acid (IPA), a gut microbiota-derived metabolite of tryptophan. IPA exhibits similar effects as its precursor, tryptophan, on energetic balance and the cardiovascular system. It also has positive impacts on a cellular level by preventing oxidative stress and inflammation. Factors such as risk factors of atherosclerosis decrease IPA synthesis, while protective factors like the Mediterranean diet tend to increase its concentration. IPA shows promise as a new target linking gut health with the cardiovascular system.

Indolepropionic Acid, a Gut Bacteria-Produced Tryptophan Metabolite and the Risk of Type 2 Diabetes and Non-Alcoholic Fatty Liver Disease

Ratika Sehgal et al.

Summary: The relationship between gut microbiota, diet, and the human body is complex, with tryptophan derivative indolepropionic acid (IPA) playing a crucial role in metabolic and immune functions and in the onset and development of metabolic disorders such as T2D and NAFLD. IPA levels are heavily influenced by diet, particularly dietary fiber, and show significant variations among individuals, which may be partially explained by genetic variants associated with specific diseases like T2D.

Microbial Metabolite 3-Indolepropionic Acid Mediates Immunosuppression

Carlos Guijas et al.

Summary: The microbial-derived metabolite 3-indolepropionic acid (3-IPA) plays an important role in T-cell exhaustion and immunosuppression. Metabolomic analysis reveals that 3-IPA can improve survival rates in mouse models infected with lymphocytic choriomeningitis virus (LCMV) Clone 13 (Cl13). This finding has significant implications for the treatment of autoimmune diseases, type I diabetes, and COVID-19.

Plasma metabolites associated with chronic kidney disease and renal function in adults from the Baltimore Longitudinal Study of Aging

Yuko Yamaguchi et al.

Summary: The study aimed to clarify the relationship of plasma metabolites with chronic kidney disease (CKD) and renal function in humans. The results showed that various plasma metabolites were associated with CKD and estimated glomerular filtration rate (eGFR), indicating alterations in multiple metabolic pathways in adults with CKD and impaired renal function.

The quantitative determination of indolic microbial tryptophan metabolites in human and rodent samples: A systematic review

George M. Anderson

Summary: This review compiled and discussed concentrations of indolic microbial metabolites of tryptophan in various human and rodent bodily fluids, aiming to compare existing reports, resolve discrepancies, draw biological inferences, survey analytical methodologies, and identify areas needing further attention. The importance of indolic compounds in human health and disease prompted this systematic review of the literature, which included key word searches of multiple databases and bibliographies of identified publications.
