Redesigning an antibody H3 loop by virtual screening of a small library of human germline-derived sequences

Summary: This study explores the replacement of the entire H3 loop in biologic therapeutics, resulting in the discovery of optimized H3 sequences from human germline-derived sources. Experimental testing showed that the re-designed H3 loops contributed positively to overall binding affinity, with some hits exhibiting similar or better affinities relative to the parental antibody. Additionally, the re-designed H3 variants demonstrated good developability attributes such as high thermal stability and no aggregation.
