The Cell Wall, Cell Membrane and Virulence Factors of Staphylococcus aureus and Their Role in Antibiotic Resistance

Philip Nikolic et al.

Summary: Antibiotic resistant strains of bacteria pose a serious threat to human health and alternative methods are needed to treat resistant bacterial infections. Methicillin resistant Staphylococcus aureus bacteria are resistant to most antibiotics, including last resort drugs. Changes in cell wall, cell membrane, and virulence factors contribute to antibiotic resistance. New antimicrobial strategies can target cell membrane lipids and virulence factors as adjuvant to traditional antibiotic therapy.

Transcriptional analysis of prsA and vraTS regulatory system in methicillin resistant Staphylococcus aureus against oxacillin stress

Deepshikha Bhowmik et al.

Summary: This study observed the transcriptional response of the prsA and vraTSR regulatory systems under oxacillin stress in Staphylococcus aureus. The results showed that the expressions of vraS and vraT increased with the increase in oxacillin concentration, while prsA did not show significant expression. This highlights the role of vraS and vraT regulatory system in conferring methicillin-resistant phenotype in S. aureus when exposed to subinhibitory concentrations of oxacillin.

Staphylococcus aureus is able to generate resistance to novel lipoglycopeptide antibiotic gausemycin A

Darya V. V. Poshvina et al.

Summary: Gausemycin A, a novel lipoglycopeptide, exhibits bactericidal activity against methicillin-resistant Staphylococcus aureus. The resistant variant S. aureus 5812R, which is 80 times more resistant compared to the parent strain, showed cross-resistance to multiple antibiotics. Whole genome sequencing revealed genetic mutations and deletions in S. aureus 5812R, as well as changes in the composition of fatty acids in the cell membrane. The combination of Gausemycin A and ciprofloxacin demonstrated a synergistic effect against S. aureus 5812R.

Global Inventory of ClpP- and ClpX-Regulated Proteins in Staphylococcus aureus

Volker C. Kirsch et al.

Summary: This study investigated the functional roles of ClpP and ClpX in controlling pathogenesis in Staphylococcus aureus, utilizing genomic deletion mutants and an inactive ClpP active-site mutant strain. The results revealed downregulation of virulence, purine/pyrimidine biosynthesis, iron uptake, and stress response in mutant strains compared to the wildtype, with implications for the development of novel virulence inhibitor classes.

Gausemycins A,B: Cyclic Lipoglycopeptides from Streptomyces sp.**

Anton P. Tyurin et al.

Summary: A novel family of natural lipoglycopeptides produced by Streptomyces sp. INA-Ac-5812, including gausemycins A and B, have been reported. These compounds exhibit pronounced activity against Gram-positive bacteria and show significant differences in mechanism compared to known glyco- and lipopeptides.

Bacterial Targets of Antibiotics in Methicillin-Resistant Staphylococcus aureus

Harshad Lade et al.

Summary: MRSA is a prevalent bacterial pathogen worldwide, and finding new antibiotic targets is crucial for successful treatment. Most antibiotics target essential biochemical processes of S. aureus, such as cell wall synthesis. Further research into novel antibiotics targeting bacterial cellular processes could lead to new therapeutic strategies against antibiotic-resistant pathogens.

Loss of the ClpXP Protease Leads to Decreased Resistance to Cell-Envelope Targeting Antimicrobials in Bacillus anthracis Sterne

Lang Zou et al.

Summary: The ClpX ATPase is crucial for Bacillus anthracis resistance to cell envelope targeting antibiotics by forming ClpXP protease. Disruption of clpP genes increases susceptibility to antimicrobials, but neither gene is essential for virulence. Changes in cell envelope morphology, including increased hydrophobicity and cell division defects, were observed in Delta clpX strain.

The phosphatidylglycerol phosphate synthase PgsA utilizes a trifurcated amphipathic cavity for catalysis at the membrane-cytosol interface

Bowei Yang et al.

Summary: Phosphatidylglycerol phosphate synthase (PgsA) plays a crucial role in pathogenic bacteria such as Staphylococcus aureus and mutations in PgsA are associated with resistance to daptomycin. The crystal structures of S. aureus PgsA reveal detailed insights into the catalytic process and provide a structural framework for the development of antimicrobial agents.
