Application of recyclable CRISPR/Cas9 tools for targeted genome editing in the postharvest pathogenic fungi Penicillium digitatum and Penicillium expansum

Sandra Garrigues et al.

Summary: In this study, we successfully developed a method for implementing CRISPR/Cas9 technology in Penicillium digitatum and Penicillium expansum fungi for the first time, targeting the wetA gene. Mutants showed altered growth, pathogenicity, conidiogenesis, germination, and sensitivity to antifungal proteins. This work demonstrates the feasibility of the CRISPR/Cas9 system and expands the genetic engineering tools available for these postharvest pathogens.

Molecular basis and regulatory mechanisms underlying fungal insecticides' resistance to solar ultraviolet irradiation

Sen-Miao Tong et al.

Summary: This review summarizes the research progress on fungal conidial UV resistance, focusing on the genetic basis of UV resistance in insect pathogen fungi, as well as the molecular mechanisms underlying photorepair and photoreactivation of UV-induced DNA lesions in conidia. The study suggests exploring a regulatory network of fungal photoprotection and improving the development and application strategies of UV-resistant fungal insecticides.

Three Small Cysteine-Free Proteins (CFP1-3) Are Required for Insect-Pathogenic Lifestyle of Metarhizium robertsii

Ya-Ni Mou et al.

Summary: This study identified a unique protein called CFP as a virulence factor in Beauveria bassiana, a main source of fungal insecticides. The study also showed the essential roles of CFP homologs in Metarhizium robertsii in cuticle infection and virulence. Transcriptomic analysis revealed dysregulation of genes involved in virulence-related processes, providing insight into genetic improvement of fungal insecticidal activity.

Roles of BrlA and AbaA in Mediating Asexual and Insect Pathogenic Lifecycles of Metarhizium robertsii

Jin-Guan Zhang et al.

Summary: This study characterizes the regulatory roles of BrlA and AbaA orthologs in Metarhizium robertsii and reveals their conserved roles in asexual developmental control as well as their indispensable roles in fungal adaptation to the insect-pathogenic lifecycle and host habitats. Furthermore, the study shows that the BrlA- or AbaA-controlled gene expression networks are largely different between the two insect pathogens, despite similar phenotypic defects in the absence of either brlA or abaA.

Identification of a key G-protein coupled receptor in mediating appressorium formation and fungal virulence against insects

Junmei Shang et al.

Summary: This study revealed the evolution of GPCR genes in the entomopathogenic Metarhizium species, showing an expansion of Pth11-like GPCRs in generalist species. Deletion of MrGpr8 resulted in failure of appressorium formation and loss of virulence during topical infection of insects. The results shed light on understanding fungus-interactions mediated by GPCRs.

Distinctive role of fluG in the adaptation of Beauveria bassiana to insect-pathogenic lifecycle and environmental stresses

Chong-Tao Guo et al.

Summary: Disruption of fluG in Beauveria bassiana leads to limited conidiation defect, which can be alleviated with longer incubation time and is associated with the up-regulation/down-regulation of other related genes. Increased sensitivities to stresses and decreased virulence were observed in Delta fluG strains, indicating the essential role of fluG in fungal adaptation to the insect-pathogenic lifecycle.
