Production and characterization of monoclonal antibodies against mandarinfish ranavirus and first identification of pyloric caecum as the major target tissue

Summary: This study successfully produced and characterized monoclonal antibodies (mAbs) against Mandarinfish ranavirus (MRV). Seven mAbs that specifically recognized MRV/LMBV were obtained through Western blotting screening and they targeted late expression structural proteins of MRV. The tissue tropism of MRV was re-assessed using immuno-histochemistry and immuno-histo-fluorescence, and it was found that spleen, kidney, intestines, and pyloric caecum are the major target tissues. Infected intestines and pyloric caecum showed enlarged, multinucleated cells with intracytoplasmic inclusions as the target cells of MRV.

A mandarinfish Siniperca chuatsi infection and vaccination model for SDDV and efficacy evaluation of the formalin-killed cell vaccine in yellowfin seabream Acanthopagrus latus

Summary: A model for studying the infection and vaccination of SDDV was established using mandarinfish. The results showed that mandarinfish is highly susceptible to SDDV infection and the virus loads were highest in the spleen, followed by other organs. Immunization with an SDDV FKC vaccine provided high levels of protection in both mandarinfish and yellowfin seabream.

Summary: A bivalent inactivated vaccine against ISKNV/RSIV and SDDV was developed and evaluated in Asian seabass. The survival rates of immunized fish were between 86.67% and 96.7%. The study also provided important information on the clinical symptoms and pathological changes caused by the viruses, contributing to the prevention and control of megalocytiviruses in Asian seabass.

A new embryonic cell line YCE1 from large yellow croaker (Larimichthys crocea) and its susceptibility to large yellow croaker iridovirus

Summary: A new cell line, YCE1, was established and characterized from Larimichthys crocea for the purpose of in vitro culture models for viral pathogens. YCE1 cells, identified as predominantly fibroblast-like, could form a monolayer at 15-37 degrees C and had an optimum growth temperature of 26 degrees C. The susceptibility test showed noticeable and stable cytopathic effects (CPEs) in the YCE1 cells infected with large yellow croaker iridovirus (LYCIV), making it a viable tool for studying host-pathogen interactions.

Whole-genome analysis of red sea bream iridovirus spread in 2021 in Japan provided epidemiological and viral traits insight

Summary: This study analysed nine whole-genome sequences of Red Sea bream iridovirus (RSIV) isolated in Oita and Ehime Prefectures in 2021. The results showed highly uniform sequences among the nine isolates, regardless of location or host species. Compared to previous isolates, RSIV isolated in 2021 was genetically different, potentially originating from a different common ancestor. Additionally, sequence mutations were found in protein-coding sequences of RSIV isolated in 2021, which may be related to viral pathogenicity and infectivity.

Identification, histopathology, and phylogenetic analysis of an iridovirus from cultivated silver pomfret in Zhejiang Province, East China

Summary: A serious disease outbreak occurred in silver pomfret at a marine fish farm in Ningbo, Zhejiang Province, China in the summer of 2018, causing high fish mortality. Infected fish showed typical signs of red sea bream iridovirus (RSIV) infection, including enlarged spleen and pale liver. Phylogenetic analysis revealed that the iridovirus from silver pomfret belongs to a subgroup of the RSIV type of the Megalocytivirus genus.

First detection of infectious spleen and kidney necrosis virus (ISKNV) associated with massive mortalities in farmed tilapia in Africa

Complete genome analysis of a red seabream iridovirus (RSIV) isolated from Asian seabass (Lates calcarifer) in India

Summary: This study reported the first case of RSIV infection in India, leading to mass mortalities of Asian seabass, Lates calcarifer. The complete genome of RSIV-LC was found to be 111,557 bp in size with 38 predicted functional proteins and 26 core genes homologous to iridovirus complete genomes. Phylogenetic analysis revealed RSIV-LC to belong to the Megalocytivirus genus of RSIV-Genotype II.

Scale Drop Disease Virus Associated Yellowfin Seabream (Acanthopagrus latus) Ascites Diseases, Zhuhai, Guangdong, Southern China: The First Description

Summary: The study describes the prevalence of Scale drop disease virus (SDDV) in farmed Asian seabass in Southeast Asia, isolates an SDDV ZH-06/20, and analyzes its genome and proteome. SDDV ZH-06/20 was found to cause 100% mortality in juvenile yellowfin seabream, characterized by severe ascites. This study provides valuable information for better understanding SDDV-associated diseases in farmed fish.
