Petabase-scale sequence alignment catalyses viral discovery

Robert C. Edgar et al.

Summary: Public databases contain a vast amount of nucleic acid sequences, but efficient methods for systematic exploration of this library have been lacking. In this study, we developed a cloud computing infrastructure, Serratus, to enable ultra-high-throughput sequence alignment at the petabase scale. By searching millions of diverse samples, we identified over 10(5) novel RNA viruses and characterized their environmental reservoirs. To facilitate viral discovery, we established a comprehensive database of these data and tools. Expanding the known sequence diversity of viruses has important implications for understanding emerging pathogens and improving pathogen surveillance.

Seroepidemiologic study on convalescent sera from dengue fever patients in Jinghong, Yunnan

Yingshuo Ma et al.

Summary: This study investigated the dynamic changes of DENV-specific antibodies and the risk of antibody-dependent enhancement (ADE) caused by secondary infection with different serotypes of dengue virus (DENV) in dengue fever patients. The results showed a gradual decrease in DENV-specific antibodies and neutralizing antibody titers after DENV infection, which may increase the risk of ADE. The risk of ADE was positively associated with the extension of convalescent phase, with higher odds ratio in DENV-4 infection. However, with the recovery period lengthened, the risk of secondary infection with DENV-1 and DENV-2 was reduced.

RNA virome diversity and Wolbachia infection in individual Drosophila simulans flies

Ayda Susana Ortiz-Baez et al.

Summary: This study investigates the potential virus protective effect of the wAu strain of Wolbachia on individual wild-caught flies, finding no significant variation in virus composition and abundance between Wolbachia-infected and uninfected flies. It remains unclear if wAu affects viral infection by increasing tolerance rather than inducing complete resistance. The study also uncovers the natural virome diversity of D. simulans, identifying known RNA viruses and discovering five novel viruses.

A Metagenomic Analysis of Mosquito Virome Collected From Different Animal Farms at Yunnan-Myanmar Border of China

Muddassar Hameed et al.

Summary: The study conducted metagenomic analysis of mosquito samples from animal farms in Yunnan province of China, revealing a highly diverse viral community with more than 19 viral taxonomic families. The viral reads included various animal viruses, suggesting a potential circulation of these viruses in vertebrate hosts. This study provides comprehensive knowledge of diverse viral populations in Yunnan province, which may pose a potential threat to both humans and domestic animals.

Metagenomic shotgun sequencing reveals host species as an important driver of virome composition in mosquitoes

Panpim Thongsripong et al.

Summary: High-throughput nucleic acid sequencing has accelerated the discovery of viruses in the environment, particularly among mosquitoes which are important for public health. Study in Thailand shows that virus presence patterns are primarily defined by host species rather than geography or habitat. Various microorganisms were also detected, emphasizing the importance of ecological information in viromic studies.

Towards an ecosystem model of infectious disease

James M. Hassell et al.

Summary: The close connection between human society and the natural environment is leading to the emergence of novel pathogens, with devastating consequences for both humans and animals. Developing more complete systems ecology models can greatly enhance our understanding of the interactions between parasites, pathogens, and the environment, ultimately improving our ability to prepare for future pandemics. This Perspective examines how integrating pathogens into systems ecology models can transform our understanding of ecosystem functioning, disease ecology, and the detection and control of zoonoses.

The virome of vector mosquitoes

Joao P. P. de Almeida et al.

Summary: Mosquitoes are major vectors for arboviruses of medical importance, with a virome in mosquitoes being of special interest. In addition to arboviruses, mosquitoes carry ISVs that may interact with arboviruses and impact mosquito vector competence. Research on the virome of vector mosquitoes may lead to novel strategies for preventing arbovirus outbreaks.

MODELING AND ANALYSIS OF THE IMPLEMENTATION OF THE WOLBACHIA INCOMPATIBLE AND STERILE INSECT TECHNIQUE FOR MOSQUITO POPULATION SUPPRESSION

Bo Zheng et al.

Summary: The successful elimination of the dengue vector Aedes albopictus on Shazai and Dadaosha islands in Guangzhou through the release of Wolbachia-infected mosquitoes in 2016 and 2017 has led to the development of mathematical guidance for designing effective mass release strategies. The key concerns in these strategies involve determining the frequency and quantity of Wolbachia-infected mosquito releases required for population suppression. Mathematically analyzing the data from the experimental releases on the islands has helped in formulating suppression models, setting release amount thresholds, and establishing conditions for population suppression based on release quantities.
