Summary: mTOR activation is associated with resistance to GC-based chemotherapy in T-ALL. In this study, it was found that the mTOR inhibitor everolimus increased ROS levels, activated NRF2, induced mitochondrial inner membrane depolarization and apoptosis of T-ALL cells. The combination of everolimus and dexamethasone had a synergistic effect on killing T-ALL cells.

Targeting mitochondrial ion channels for cancer therapy

Summary: Targeting mitochondrial ion channels with drugs to eliminate cancer cells is a promising approach due to differential expression and/or regulation of these channels in cancer cells compared to healthy cells. Modulation of ion channels can indirectly affect oxidative phosphorylation, which may be crucial for cancer and cancer stem cell survival. Additionally, modulation of mitochondrial ion channels leading to cytochrome c release may be advantageous in overcoming drug resistance.

Exploiting the metabolic energy demands of drug efflux pumps provides a strategy to overcome multidrug resistance in cancer

Summary: The study demonstrates that the addition of verapamil to resistant cells results in a significant reduction in ATP levels, disrupting energy homeostasis. Even in the absence of verapamil, P-gp expressing cells exhibit near maximal rates of glycolysis and oxidative phosphorylation, preventing an adequate response to the demand for ATP. Moreover, the perpetually high rate of oxidative phosphorylation in the presence of verapamil leads to elevated levels of reactive oxygen species that affect cell survival and highlight collateral sensitivity.
