Mitochondrial ATP synthase c-subunit leak channel triggers cell death upon loss of its F1 subcomplex

Summary: In this study, the c-ring of mitochondrial ATP synthase was purified from both eukaryotic and prokaryotic hosts. It was found that the purified c-ring forms a multi-conductance, voltage-gated ion channel. The addition of the ATP synthase F-1 subcomplex inhibits the channel activity, while dissociation of F-1 from F-O occurs during cell death.

Mitochondrial uncouplers induce proton leak by activating AAC and UCP1

Summary: Mitochondria generate heat through H+ leakage mediated by UCP1 and AAC. Protonophores induce H+ leakage, but their clinical application is limited. This study directly measured the effect of protonophores on H+ leakage and found that it is dependent on AAC and UCP1. Through molecular structure analysis and mathematical modeling, a mechanism of uncoupler-dependent H+ leakage through AAC is proposed.

Summary: This study investigates the inhibitory effect of kCh on mitochondrial respiratory complex I. Results suggest that kCh interacts with the ubiquinone-binding center of the enzyme and selectively inhibits the activity of certain substrates. The analogs of kCh do not exhibit similar effects, highlighting the specificity of kCh on mitochondrial function.

Summary: ATP synthase can utilize both Delta psi(m)-driven H+- and K+-transport to synthesize ATP under physiological conditions. The presence of K+ increases ATP synthesis rate in isolated mitochondria while also increasing oxygen consumption rate. The data obtained from purified F1Fo single molecule experiments are consistent with the functional data observed in intact mitochondria.

Research suggests that adenine nucleotide translocase (ANT) and other mitochondrial proteins are involved in DNP's proton leakage activity. Specifically, the crucial role of arginine 79 of ANT1 in the DNP-mediated increase of membrane conductance has been demonstrated.
