Summary: Intervertebral disc degeneration (IDD) is the pathological basis of disc degenerative diseases (DDD), characterized by reduction in the number of cells and degeneration of the extracellular matrix in the nucleus pulposus. An injectable ECM hydrogel combined with stem cell transplantation shows promise, but lacks sufficient biomechanics and stability. A genipin cross-linked decellularized nucleus pulposus hydrogel (GDH) was developed as a delivery system, showing good mechanical properties, biocompatibility, and differentiation induction in vitro. In vivo studies demonstrated that GDH loaded with adipose-derived mesenchymal stem cells (GDHA) improved stem cell survival and intervertebral disc regeneration.
