A single-cell census of mouse limb development identifies complex spatiotemporal dynamics

Summary: Through single-cell RNA sequencing of developing mouse forelimb, we identify three progenitor states: naive, proximal, and autopodial, with Msx1 as a marker for naive progenitors. Lineage tracing confirms the role of naive progenitors and locates them at the outer margin of the limb along the anterior-posterior axis. Sequential pulse-chase experiments show that the progressive transition of Msx1+ naive progenitors into proximal and autopodial progenitors coincides with their differentiation into Sox9+ chondroprogenitors, occurring along all the forming skeletal segments. Hence, the skeleton forms progressively in a complex pattern.

Gradient to sectioning CUBE workflow for the generation and imaging of organoids with localized differentiation

Summary: Advancements in organoid culture have led to various in vitro mini-organs that mimic native tissues. However, generating complex organoids with body axis patterning and maintaining their orientation is still challenging. In this study, we present a workflow using a CUBE culture device to culture organoids with morphogen gradient and section samples with the CUBE to retain gradient direction information. Our results demonstrate that hiPSC spheroids cultured with different differentiation media on opposing ends of the CUBE show localized expressions of differentiation markers, unlike the control group with homogeneous marker distribution. This workflow provides researchers with a convenient tool to generate complex organoids and study their developmental processes in vitro.
