Rutin prevents tau pathology and neuroinflammation in a mouse model of Alzheimer's disease

Summary: The study demonstrated that rutin could inhibit tau aggregation and tau oligomer-induced cytotoxicity, reduce the production of proinflammatory cytokines, protect neuronal morphology from toxic tau oligomers, and promote microglial uptake of extracellular tau oligomers. In a Tau-P301S mouse model, rutin also reduced pathological tau levels, regulated tau hyperphosphorylation, suppressed gliosis and neuroinflammation, prevented microglial synapse engulfment, and rescued synapse loss, resulting in a significant improvement of cognition. These findings suggest that rutin is a promising drug candidate for Alzheimer's disease treatment by targeting both tau and amyloid beta pathology.

Bioinformatic analysis for potential biological processes and key targets of heart failure-related stroke

Summary: This study aimed to uncover the underlying mechanisms and promising intervention targets of heart failure (HF)-related stroke through weighted gene co-expression network analysis (WGCNA). The study identified key genes involved in protein ubiquitination, Wnt signaling, and exosomes in both HF- and stroke-related key modules. OTU deubiquitinase with linear linkage specificity (OTULIN) and nuclear factor interleukin 3-regulated (NFIL3) were determined to be the key genes in HF-related stroke, contributing to the regulation of processes such as protein ubiquitination and Wnt signaling. OTULIN and NFIL3 were validated to be differentially expressed in all HF and stroke groups, suggesting their potential as promising intervention targets.
