Extracellular Vesicle Secretion by Leukemia Cells In Vivo Promotes CLL Progression by Hampering Antitumor T-cell Responses

Ernesto Gargiulo et al.

Summary: This study investigated the interactions between cancer cells and the microenvironment using mouse models of chronic lymphocytic leukemia (CLL). It found that small extracellular vesicles (sEVs) derived from CLL tissue contain specific miRNA and immune-checkpoint ligands, which alter the function and exhaustion of CD8+ T cells. Blocking sEV biogenesis delayed CLL pathogenesis and high expression of sEV-related genes correlated with poor outcomes in CLL patients.

Extracellular vesicles released by non-small cell lung cancer cells drive invasion and permeability in non-tumorigenic lung epithelial cells

Humna Hasan et al.

Summary: Extracellular vesicles (EVs) released from non-small cell lung cancer (NSCLC) cells play a crucial role in promoting cancer progression. There are differences in the secretion profile and the ability to induce phenotypic changes in non-tumorigenic cells among EVs derived from different NSCLC cell lines. EV-encapsulated RNA (EV-RNA) is identified as a key factor in mediating these phenotypes. The aggressiveness of NSCLC subtypes may be correlated with the ability of their respective EVs to induce cancerous phenotypes.

Biomimetic nanoparticles directly remodel immunosuppressive microenvironment for boosting glioblastoma immunotherapy

Tingting Wang et al.

Summary: This study demonstrates the significant improvement of immunotherapy efficacy for glioblastoma (GBM) by modulating the tumor immunosuppressive microenvironment (TIME) through novel all-in-one biomimetic nanoparticles. By remodeling the TIME, these nanoparticles activate immune responses, decrease tumor infiltration, and reduce the expression of PD-L1 on tumor cells. Modulating the TIME shows potential as a significant strategy to enhance immunotherapy for GBM and other cold tumors.

Targeting extracellular matrix stiffness and mechanotransducers to improve cancer therapy

Yangfu Jiang et al.

Summary: The cancer microenvironment plays a critical role in tumorigenesis and cancer progression. The interaction between the extracellular matrix (ECM) and tumor and stromal cells contributes to cancer cell proliferation, migration, invasion, angiogenesis, and immune evasion. Both ECM stiffness itself and ECM stiffening-induced mechanical stimuli can activate cell membrane receptors and mechanosensors, thereby modulating the malignant phenotype of tumor and stromal cells. Understanding how ECM stiffness regulates tumor progression is crucial for the development of new therapeutics.

Summary: Tumor-derived extracellular vesicles and soluble molecules can promote the differentiation of monocytes into different types of macrophages. While the vesicles specifically promote proinflammatory macrophages, the soluble molecules direct the differentiation of a specific macrophage subset through certain activation pathways. The presence of macrophages expressing the vesicle-induced signature in breast cancer patients' TAMs is associated with T cell infiltration and extended patient survival.

CD9 mediates the uptake of extracellular vesicles from cancer-associated fibroblasts that promote pancreatic cancer cell aggressiveness

Jeremy Nigri et al.

Summary: In pancreatic ductal adenocarcinoma (PDAC), signaling from stromal cells through extracellular vesicles (EVs) is involved in metastatic progression. This study identified CD9 as a key component of EVs derived from cancer-associated stromal fibroblasts (CAFs), which mediate the cross-talk between tumor cells and the stroma. Blocking CD9 impaired the uptake of EVs into PDAC cells, and CD9(+) EVs induced MAPK pathway activation, cell migration, and EMT. Inhibiting CD9 or p38 MAPK signaling impaired EV-induced cell migration and EMT. Bioinformatic analysis showed that CD9 abundance is an independent prognostic marker for poor prognosis in PDAC patients.

CD36-mediated metabolic crosstalk between tumor cells and macrophages affects liver metastasis

Ping Yang et al.

Summary: Our study reveals that the scavenger receptor CD36 is upregulated in metastasis-associated macrophages (MAMs) and deletion of CD36 in MAMs attenuates liver metastasis in mice. MAMs contain more lipid droplets and have the unique capability to engulf tumor cell-derived long-chain fatty acids carried by extracellular vesicles. These lipid-enriched vesicles are preferentially partitioned into macrophages via CD36, fueling their tumor-promoting activities and creating a highly immunosuppressive tumor microenvironment (TME) in liver metastasis.
