Restoring glutamate receptor signaling in pancreatic alpha cells rescues glucagon responses in type 1 diabetes

Julia K. Panzer et al.

Summary: This study identifies a defect in autocrine signaling in pancreatic alpha cells of individuals with type 1 diabetes, which leads to inadequate glucagon response and susceptibility to hypoglycemia. Reactivating residual AMPA/kainate receptor function with positive allosteric modulators can restore normal glucagon secretion and prevent hypoglycemia, providing a potential therapeutic approach for managing diabetes.

Glucagon Resistance and Decreased Susceptibility to Diabetes in a Model of Chronic Hyperglucagonemia

Nadejda Bozadjieva Kramer et al.

Summary: The elevation of glucagon levels and increase in alpha-cell mass in diabetes are associated with states of hyperglycemia. Activating nutrient signaling by conditional deletion of the mTORC1 inhibitor, TSC2, in alpha-cells can induce chronic hyperglucagonemia, leading to improvements in glucose homeostasis through glucagon resistance in the liver. The resistance is characterized by reduced expression of glucagon receptor (GCGR), PEPCK, and genes involved in amino acid metabolism and urea production.

Gq signaling in α cells is critical for q maintaining euglycemia

Liu Liu et al.

Summary: This study demonstrates that by activating G(q) signaling pathway, glucagon can be released to improve insulin secretion and glucose tolerance. Chronic activation of this pathway also benefits glucose metabolism in obese mice. Additionally, the activation of V1bR receptor stimulates glucagon release, highlighting its role in regulating glucagon secretion under hypoglycemic conditions.
