High-resolution 3D printing of angle-ply annulus fibrosus scaffolds for intervertebral disc regeneration

Summary: In this study, high-resolution polycaprolactone (PCL) scaffolds were successfully fabricated using a new electrohydrodynamic 3D printing technique, allowing for accurate simulation of the native annulus fibrosus (AF) structure. In vivo experiments showed that the tissue-engineered intervertebral discs (TE-IVDs) constructed with the 3D printed AF scaffold maintained disc height, reduced loss of nucleus pulposus (NP) water content, and partially restored the biomechanical function of the intervertebral disc.

Crystal Growth of 3D Poly(ε-caprolactone) Based Bone Scaffolds and Its Effects on the Physical Properties and Cellular Interactions

The results show that the scaffolds produced using these techniques exhibit distinct physiochemical properties. Melt-printed scaffolds have stronger mechanical properties, while solvent-printed scaffolds have a rougher surface, higher degradation rate, and faster stress relaxation. These differences are attributed to the different crystal growth kinetics and result in different cell-scaffold interactions.

This study demonstrates the use of embedded three-dimensional (EMB3D) printing to create multi-material polymer lattices, enabling the fabrication of a wide range of periodic and stochastic lattices with tailored mechanical responses by printing multiple materials with different mechanical properties.

Simulated stress mitigation strategies in embedded bioprinting

Embedded ink writing (EIW) is an extrusion-based printing technique that allows for low-viscosity inks, but may result in shape defects. Computational fluid dynamics simulations show that using a conical nozzle, increasing the nozzle diameter, decreasing the print speed, and decreasing the ink viscosity can improve cell viability for stress magnitude-sensitive cells, but may increase damage and shape defects for stress duration-sensitive cells.
