Comprehensive Analysis of Major Latex-Like Protein Family Genes in Cucumber (Cucumis sativus L.) and Their Potential Roles in Phytophthora Blight Resistance

Summary: In this study, 37 putative cucumber MLP genes were identified and classified into three groups. It was found that only tandem duplication occurred in the evolution of these genes. The promoter region of these CsMLP genes contained multiple regulatory cis-elements related to stress, hormone, light, and growth response, suggesting their involvement in various biological processes. Transcriptome analysis revealed a reprogramming of MLPs expression in response to Phytophthora melonis infection. Knockdown and overexpression experiments demonstrated the involvement of CsMLP1 and CsMLP5 in disease tolerance and lesion expansion. Overall, this study provides insights into the function and evolutionary relationship of MLP genes in cucumber.

Genome-wide characterization of NBS-LRR family genes and expression analysis under powdery mildew stress in Lagenaria siceraria

Summary: In this study, we investigated the NBS-LRR genes in bottle gourd and identified 84 genes categorized into seven subfamilies. Phylogenetic and motif analysis revealed two major clusters of NBS-LRR genes with TIR or coiled-coil domains. We also analyzed duplication events, motif sequences, orthologous gene clusters, protein-protein interactions, and cis-elements. Furthermore, we constructed protein structures and found a significantly up-regulated gene, Lsi04g015960, with the powdery mildew8 (RPW8) domain in PM-resistant variety. This research provides valuable insights into the NBS-LRR family genes in bottle gourd and identifies a potential candidate gene for PM tolerance breeding.

CRISPR/Cas9 based mlo-mediated resistance against Podosphaera xanthii in cucumber (Cucumis sativus L.)

Summary: This study investigates the mlo-based powdery mildew resistance mechanism in cucumber. CRISPR/Cas9 technology was used to generate loss-of-function mutants for CsaMLO1, CsaMLO8, and CsaMLO11 genes in cucumber, and these mutants were validated. The results show that CsaMLO genes associated with PM-susceptibility act as negative regulators in different defense mechanisms at different stages of infection. CsaMLO8 mutation-based resistance is linked to pre-invasive response, while CsaMLO1 and CsaMLO11 may serve as negative regulators in post-invasive defense response.

Grafting improves growth and nitrogen-use efficiency by enhancing NO3- uptake, photosynthesis, and gene expression of nitrate transporters and nitrogen metabolizing enzymes in watermelon under reduced nitrogen application

Summary: This study found that grafting with bottle gourd rootstock can improve the nitrogen use efficiency and growth performance of watermelon under reduced nitrate application. The rootstock-grafted plants showed better root morphology, NO3- accumulation, photosynthesis, and nitrogen use efficiency compared to self-grafted plants under reduced nitrate conditions. The up-regulation of nitrate transporter and nitrogen-metabolizing enzyme genes in the rootstock-grafted plants contributed to the enhanced NO3- uptake and utilization. Grafting with bottle gourd rootstock may be beneficial for the efficient production of watermelon and economical application of nitrogen fertilizer.

Comparative transcriptomic analysis of powdery mildew resistant and susceptible melon inbred lines to identify the genes involved in the response to Podosphaera xanthii infection

Summary: This study conducted RNA-seq analysis of resistant and susceptible melons to powdery mildew (PM) infection and identified significant differentially expressed genes (DEGs) related to PM resistance. The study revealed the different response and regulatory network between resistant and susceptible melons to PM infection. The study also found that genes related to phenylpropanoid biosynthesis and encoding glucan endo-1,3-beta- glucosidases and cytochrome P450 proteins may play important roles in the resistance to P. xanthii infection.

Summary: Vegetable grafting is recognized as an effective and sustainable plant production alternative. This study investigates the effects of rootstocks on scion performance and the influences of scions on root traits. Successfully grafted scions increase the ratio of root to whole plant dry matter and the ratios of root length to whole plant dry matter and to plant leaf area. Grafting with different rootstock species also affects the enzymatic activities and nutrient acquisition capacity of grafted plants.

Comparative transcriptome analysis of melon (Cucumis melo L.) reveals candidate genes and pathways involved in powdery mildew resistance

Development of KASP Markers and Identification of a QTL Underlying Powdery Mildew Resistance in Melon (Cucumis melo L.) by Bulked Segregant Analysis and RNA-Seq
