Cancer statistics, 2022

Rebecca L. Siegel et al.

Summary: Each year, the American Cancer Society compiles data on cancer occurrence and outcomes in the United States. The latest data shows that breast and prostate cancer progress has stagnated, while lung cancer has shown improvements in survival rates. Lung cancer incidence for advanced disease has declined while localized-stage rates have increased, resulting in higher survival rates. Mortality patterns align with incidence trends, with lung cancer deaths declining rapidly, breast cancer deaths slowing, and prostate cancer deaths stabilizing.

FBXO2 targets glycosylated SUN2 for ubiquitination and degradation to promote ovarian cancer development

Jing Ji et al.

Summary: The study reveals that FBXO2 acts as an E3 ubiquitin ligase and promotes the degradation of SUN2 protein, which is associated with worse prognosis in ovarian cancer. The transcription factor SOX6 enhances the expression of FBXO2 by recognizing a response element on its promoter. Abnormally high expression of FBXO2 leads to degradation of SUN2 protein, promoting cell proliferation and ultimately contributing to the progression of ovarian cancer.

FBXL6 depletion restrains clear cell renal cell carcinoma progression

Yongchun Yu et al.

Summary: In this study, a survival model based on the F-box gene signature was established to evaluate the prognosis of patients with ccRCC. FBXL6 was identified as a diagnostic and prognostic marker in ccRCC. In vitro and in vivo experiments demonstrated that the depletion of FBXL6 inhibited cell proliferation and induced apoptosis in ccRCC cells.

FBXO39 predicts poor prognosis and correlates with tumor progression in cervical squamous cell carcinoma

Yanru Yang et al.

Summary: The study found that FBXO39 is highly expressed in cervical squamous cell carcinoma and is closely related to patient prognosis. Further experiments confirmed that overexpression of FBXO39 promotes the proliferation, migration, and invasion of cancer cells. Experimental results on C-33A cells showed that downregulation of FBXO39 can inhibit tumor growth. These findings suggest that FBXO39 may serve as a therapeutic target for cervical squamous cell carcinoma (CESC).

Therapeutic strategies for gastric cancer targeting immune cells: Future directions

Yan Zhao et al.

Summary: Gastric cancer is a highly prevalent and lethal malignancy, and immunotherapy has shown survival benefits for GC patients. Targeted immune cells have emerged as a promising therapeutic strategy in the field of oncology. This review discusses the role and significance of immune cells in the tumor microenvironment of GC and summarizes the current landscape of immunotherapy, including immune checkpoint inhibitors and adoptive cell therapy. While more clinical data is needed, targeting immune cells provides a new direction for GC immunotherapy.

A Bayesian network meta-analysis on systemic therapy for previously treated gastric cancer

Jung Han Kim et al.

Summary: A systematic literature review identified 20 phase III randomized controlled trials on salvage treatment of advanced gastric cancer, with olaparib plus paclitaxel, paclitaxel plus ramucirumab, and pembrolizumab showing potential as the most effective regimens. Moreover, nivolumab, chemotherapy, and apatinib demonstrated significant overall survival benefit compared to best supportive care for third-line treatment. In conclusion, pembrolizumab may be preferable for PD-L1-expressing AGC in the second-line setting, while paclitaxel-based combinations are recommended for PD-L1-negative AGC, and nivolumab may be the preferred third-line treatment.

FBXL6 degrades phosphorylated p53 to promote tumor growth

Yajun Li et al.

Summary: This study identified the amplified and over-expressed FBXL6 in colorectal cancer patients, which is highly correlated with poor prognosis, and revealed its mechanism in suppressing p53 signaling by targeting phospho-p53 for degradation. The feed-forward loop of FBXL6-p53, where p53 transcriptionally suppresses FBXL6 expression, might serve as a potential therapeutic target for CRC treatments.

HOXA10 mediates epithelial-mesenchymal transition to promote gastric cancer metastasis partly via modulation of TGFB2/Smad/METTL3 signaling axis

Chenlong Song et al.

Summary: Our study demonstrated that HOXA10 is upregulated in gastric cancer, especially in patients with lymph node metastasis. Functionally, HOXA10 promotes migration and invasion of gastric cancer cells, as well as accelerates lung metastasis in vivo. The EMT process mediated by the HOXA10/TGFB2/Smad/METTL3 signaling axis plays a crucial role in gastric cancer progression and metastasis.

Zeynep Ozman et al.

Summary: The study demonstrated that valproic acid (VPA) induces epithelial-to-mesenchymal transition (EMT) and cell migration in triple negative breast cancer cells by regulating the expression levels of Snail, E-cadherin, AKT, and GSK3 beta.

Tenascin-c knockdown suppresses vasculogenic mimicry of gastric cancer by inhibiting ERK- triggered EMT

Xing Kang et al.

Summary: The study found that Tenascin-c (TNC) is upregulated in gastric cancer tissue and is associated with vasculogenic mimicry (VM) formation and poor prognosis. Inhibition of TNC can significantly suppress VM formation and cell proliferation in gastric cancer. Targeting TNC and ERK may be a potential therapeutic approach to inhibit gastric cancer growth and metastasis.

mRNA Expression Analysis of E-Cadherin, VEGF, and MMPs in Gastric Cancer: a Pilot Study

Puneet Kumar et al.

Summary: This study investigated the mRNA expression of VEGF, E-cadherin, and MMPs in gastric cancer tissues in the Indian population. Results showed differences in the expression levels of these genes compared to control tissues. Analysis was done on the diagnostic utility and association of these genes with clinicopathological characteristics and prognosis.
