Emerging nanomedicines of paclitaxel for cancer treatment

Qin Chen et al.

Summary: Malignant tumor remains a major threat to human health. Although targeted therapeutic strategies have developed rapidly, treatments that act on a single target are prone to tumor resistance. Therefore, traditional broadspectrum chemotherapy still plays a critical role in clinical cancer therapy. However, most chemotherapeutic drugs have unsatisfactory therapeutic efficacy and severe off-target toxicity. To overcome this, researchers have focused on developing novel nano-formulations for efficient delivery of chemotherapeutic drugs.

Minor change in the length of carbon chain has a great influence on the antitumor effect of paclitaxel-fatty alcohol prodrug nanoassemblies: Small roles, big impacts

Xin Wang et al.

Summary: Prodrug-based nanoassembly with carbon chains of different lengths (C-8, C-10, C-12) was found to enhance self-assembly and drug delivery efficiency, leading to improved antitumor efficacy in the optimal paclitaxel-dodecanol prodrug nanoassemblies compared to Taxol and Abraxane. These findings are significant for the rational design of advanced nanomedicines for cancer therapy.

In-Situ Implant Formulation of Laurate and Myristate Prodrugs of Dolutegravir for Ultra-Long Delivery

Tahir Khuroo et al.

Summary: The present work focuses on synthesizing prodrugs of dolutegravir (DTG) for ultra-long delivery purpose. The prodrugs were characterized and found to lower the melting point and solubility of DTG while increasing its lipophilicity. Furthermore, the prodrugs and drugs were formulated into implants to achieve prolonged drug release.

Trends in advanced oral drug delivery system for curcumin: A systematic review

Suchiwa Pan-On et al.

Summary: Although curcumin has health benefits, its clinical application is limited by its poor solubility and stability. Nanocarrier-based drug delivery systems (NDS) are effective in improving curcumin's physicochemical and pharmacological effects. This study reviewed evidence of lipid and polymer-based NDS for oral delivery of curcumin. Most studies found that NDS increased curcumin absorption compared to free curcumin.

σ-Conjugation and H-Bond-Directed Supramolecular Self-Assembly: Key Features for Efficient Long-Lived Room Temperature Phosphorescent Organic Molecular Crystals

Catherine Demangeat et al.

Summary: Long-lived room temperature phosphorescence from organic molecular crystals is achieved by designing two isomeric molecular phosphors, highlighting the critical role of the two degrees of freedom of the sigma-conjugated bridge on the chromophore optical properties. The crystal structures of the efficient phosphorescent materials demonstrate an unprecedented well-organization of the emitters into a 2D rectangular columnar-like supramolecular structure stabilized by intermolecular H-bonding.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Small-Molecule Prodrug Nanoassemblies: An Emerging Nanoplatform for Anticancer Drug Delivery

Guanting Li et al.

Summary: SMP-NAs, formed by the self-assembly of prodrugs, have been widely investigated for their ultrahigh drug-loading and minimal adverse reactions, offering potential for improved antitumor efficiency compared to traditional nanomedicines. They are being used in chemotherapy, phototherapy, immunotherapy, and tumor diagnosis, and their co-delivery capabilities can address the challenges of tumor heterogeneity and multidrug resistance. Smart stimuli-responsive SMP-NAs and combination therapies based on SMP-NAs are also highlighted, emphasizing the structure-function relationships.
