Efficient delivery of VEGF-A mRNA for promoting diabetic wound healing via ionizable lipid nanoparticles

Wenhui Zha et al.

Summary: Diabetes often results in chronic non-healing wounds, and VEGF-A is important for their treatment. However, the use of VEGF-A protein is limited, so this study focuses on using VEGF-A mRNA to accelerate wound healing. The researchers developed LNP/VEGF-A mRNA nanoparticles and demonstrated their efficacy in promoting wound healing in vitro and in vivo.

Ionizable lipid nanoparticles deliver mRNA to pancreatic ? cells via macrophage-mediated gene transfer

Jilian R. Melamed et al.

Summary: Systemic delivery of mRNA to organs outside the liver, spleen, and lungs is difficult. This study presents a novel strategy using lipid nanoparticles to deliver mRNA specifically to the pancreas. Intraperitoneal administration of lipid nanoparticles containing cationic helper lipids results in robust and specific protein expression in the pancreas, with a majority occurring in insulin-producing beta cells. The delivery of pancreatic mRNA is found to be dependent on horizontal gene transfer by peritoneal macrophage exosome secretion. This strategy holds potential for gene therapies targeting pancreatic diseases such as diabetes and cancer.

Incorporation of curcumin-loaded lipid-based nano delivery systems into food: Release behavior in food simulants and a case study of application in a beverage

Raquel F. S. Goncalves et al.

Summary: Curcumin-loaded lipid-based nano delivery systems (nanoemulsions-NE, solid lipid nanoparticles-SLN and nanostructured lipid carriers-NLC) were evaluated for in vitro release kinetics and particle stability under different food simulants. Their physicochemical properties in a model beverage during storage were also assessed. The bioaccessibility of curcumin in beverages containing the nano delivery systems was determined through an in vitro digestion process. The release of curcumin was higher and particle stability was lower in lipophilic food simulants compared to hydrophilic simulants. NLC and SLN showed good particle stability in the beverage, while NE showed high instability. NLC had a slightly higher stability in terms of color and SLN had higher curcumin bioaccessibility but lower stability.

Lipid Nanoparticles Deliver the Therapeutic VEGFA mRNA In Vitro and In Vivo and Transform Extracellular Vesicles for Their Functional Extensions

Muhammad Nawaz et al.

Summary: The study demonstrates that LNPs can deliver VEGF-A mRNA to cells, and a fraction of the internalized mRNA is secreted via EVs. EVs derived from cardiac progenitor cells are the most efficient in promoting angiogenesis. Additionally, the delivery of VEGF-A mRNA via EVs or LNPs results in locally produced protein without spillover to the liver and minimal production of inflammatory cytokines in cardiac tissue.

Glioblastoma multiforme targeted delivery of docetaxel using bevacizumab-modified nanostructured lipid carriers impair in vitro cell growth and in vivo tumor progression

Leonardo Delello Di Filippo et al.

Summary: This study aimed to evaluate the effect of bevacizumab-functionalized docetaxel-loaded nanostructured lipid carriers (BVZ-NLC-DTX) against glioblastoma multiforme (GBM) using in vitro and in vivo models. The results showed that BVZ-NLC-DTX selectively increased the cytotoxicity of docetaxel in GBM cells and promoted cell death by apoptosis. In the in vivo experiment, BVZ-NLC-DTX significantly reduced tumor volume. Therefore, this study contributes to the development of more efficient nanocarriers for improving the efficacy of GBM drug treatment.

Claudia Viegas et al.

Summary: Lipid nanoparticles are effective carriers for delivering therapeutic proteins, providing biocompatibility and biodegradability. Different lipid carriers, such as liposomes, solid lipid nanoparticles, and nanostructured lipid carriers, can be used to load therapeutic proteins. Among these, nanostructured lipid carriers are gaining popularity due to their high loading capacity and sustained release. The production methods need to consider both the desired features of the nanocarriers and the maintenance of the protein structure and bioactivity.
