NF-kappa B as a regulator of cancer metastasis and therapy response: A focus on epithelial-mesenchymal transition

Sepideh Mirzaei et al.

Summary: Metastasis of tumor cells is a significant challenge in cancer patients, and the activation of nuclear factor-kappaB (NF-kappa B) plays a crucial role in regulating the epithelial-to-mesenchymal transition (EMT). The activation of NF-kappa B enhances the invasive properties of tumor cells through inducing EMT mechanism, and this has been confirmed in various cancers.

Neera Yadav et al.

Summary: This study demonstrates the potential of PLGA-QNPs in anticancer therapy. PLGA-QNPs significantly reduce the viability of cervical and breast cancer cells, induce apoptosis, and suppress cancer progression through regulation of gene expression and signaling pathways. In a mammary adenocarcinoma rat model, PLGA-QNPs also reduce tumor burden and prolong tumor latency. Therefore, PLGA-QNPs may serve as an effective drug delivery system for anticancer therapy.

Lineage plasticity in prostate cancer depends on JAK/STAT inflammatory signaling

Joseph M. Chan et al.

Summary: Drug resistance in cancer is often associated with tumor cell lineage plasticity. The molecular mechanisms driving this plasticity have not been fully understood. This study shows that prostate cancer plasticity is initiated in a population of cells with mixed luminal-basal phenotype and is dependent on increased JAK and FGFR activity. Single-cell analysis in patients with metastatic disease confirms the presence of mixed-lineage cells with increased JAK/STAT and FGFR signaling.

Autophagy, ferroptosis, pyroptosis, and necroptosis in tumor immunotherapy

Weitong Gao et al.

Summary: Immunotherapy has made significant breakthroughs in cancer treatment, but the poor response of some tumors limits its application. Inducing non-apoptotic regulated cell death (RCD) may convert these unresponsive tumors into those that respond to immunotherapy. Autophagy, ferroptosis, pyroptosis, and necroptosis have been found to have both inhibitory and synergistic effects on antitumor immune responses, suggesting that targeted therapies against these processes combined with immunotherapy may be effective even in resistant tumors.

Michela Marsico et al.

Summary: Metabolic reprogramming is crucial for cancer cells to meet high energy needs and maintain redox balance, with glutamine metabolism playing a key role. In the study of hepatocellular carcinoma, up-regulation of GDH1 was observed, and inhibition of this enzyme showed promising results in reducing cell proliferation and identifying potential inhibitors. These findings suggest that targeting hGDH1 could be a viable strategy to disrupt the metabolic reprogramming of HCC cells.

Quercetin as a protective agent for liver diseases: A comprehensive descriptive review of the molecular mechanism

Xingtao Zhao et al.

Summary: Quercetin, as a major flavonoid compound, exhibits good pharmacological activities in treating liver diseases by influencing multiple targets and pathways, including anti-fat accumulation, anti-inflammatory, and antioxidant effects. It inhibits inflammation mainly through NF-kappa B/TLR/NLRP3, reduces oxidative stress mediated by PI3K/Nrf2, and affects autophagy activation by mTOR. The compound also shows different mechanisms of action at different stages of liver diseases, targeting factors related to PPAR, UCP, and PLIN2 in steatosis, and inhibiting stromal ECM deposition in fibrosis, among others.
