Nanoparticles of N-Vinylpyrrolidone Amphiphilic Copolymers and Pheophorbide a as Promising Photosensitizers for Photodynamic Therapy: Design, Properties and In Vitro Phototoxic Activity

Summary: A series of nanoparticles with hydrodynamic radius ranging from 20 to 100 nm were developed by solubilizing a hydrophobic dye using amphiphilic copolymers. The photophysical properties and biological activity of the nanoparticles were studied, and it was found that the aggregation state of the dye is reduced and the singlet oxygen quantum yield and fluorescence signal increase upon interaction with biological membranes or tissue homogenates. The nanoparticles showed a higher phototoxic effect on HeLa cells compared to a reference dye, suggesting their potential for targeted drug delivery.

Design of A3B-Porphyrin Conjugates with Terpyridine as Potential Theranostic Agents: Synthesis, Complexation with Fe(III), Gd(III), and Photodynamic Activity

Summary: This paper presents the design and synthesis of new multifunctional porphyrin-based therapeutic agents, which have potential applications in therapy and diagnosis. The zinc complexes of A3B-type meso-arylporphyrins were modified with chelating ligands based on 4'-(4-methylphenyl)-2,2':6',2''-terpyridine derivatives to obtain novel complexes with Gd(III) and Fe(III). The aggregation behavior, photophysical properties, and photochemical properties of these porphyrins were studied, and the micellar Pluronic F127 formulation was found to be the most effective solubilizer. In vitro cytotoxicity studies showed that the compounds had significant toxic effects under irradiation, but no toxicity without irradiation.

The review also examines the properties of new photosensitizers, their photoactivity, the generation of singlet oxygen, and the potential use of targeted photodynamic therapy in clinical practice.

Summary: Tetraphenylporphyrin (H2TPP) and zinc tetraphenylporphyrin (ZnTPP) are widely used benchmark molecules with rich visible-region spectra and broad structural analogy to chlorophylls, but there is a significant variability in literature values for key photophysical parameters. A comprehensive review of 871 publications from 1940 to September 2020 found that the choice of values for parameters such as molar absorption coefficient, fluorescence quantum yield, and singlet excited-state lifetime can have far-reaching consequences in photochemistry applications. Different instrumentation and sources of error contribute to the wide range of reported values for these parameters.
