HuMSC-EV induce monocyte/macrophage mobilization to orchestrate neovascularization in wound healing process following radiation injury

Celine Loinard et al.

Summary: This study investigates the mechanisms by which human mesenchymal stem cell-derived extracellular vesicles (HuMSC-EV) promote angiogenesis and neovascularization after radiation injury. The study found that HuMSC-EV improved neovascularization of irradiated tissue by stimulating angiogenesis, increasing capillary density, and producing proangiogenic factors. HuMSC-EV also promoted vasculogenesis by recruiting and differentiating bone marrow progenitors. Furthermore, HuMSC-EV enhanced arteriogenesis by mobilizing monocytes and promoting their recruitment to the injured site. These findings suggest that HuMSC-EV could be a potential alternative to stem cells for therapeutic neovascularization in tissue repair.

Uptake, functionality, and re-release of extracellular vesicle-encapsulated cargo

Killian O'Brien et al.

Summary: Extracellular vesicles (EVs) are membrane-encapsulated particles carrying genetically active and protein/lipid cargo that can impact recipient cell function. This study demonstrates the functional delivery of EV-encapsulated RNA and protein cargo using luminescence and fluorescence reporters. The research highlights the retention of internalized cargo in the endosomal compartment and the re-release of intact EVs by recipient cells. These findings shed light on critical factors in cargo delivery and enhance the therapeutic potential of EVs.

Human Umbilical Cord Mesenchymal Stem Cell-Derived Exosomes Attenuate Myocardial Infarction Injury via miR-24-3p-Promoted M2 Macrophage Polarization

Feng Zhu et al.

Summary: Exosomes derived from human umbilical cord mesenchymal stem cells (UMSC-Exos) have been found to improve cardiac function by promoting M2 macrophage polarization and reducing excessive inflammation. Exosomal miR-24-3p acts as a mediator by inhibiting Plcb3 expression and NF-kappa B pathway activation to promote M2 macrophage polarization.

cAMP levels regulate macrophage alternative activation marker expression

Swamy Polumuri et al.

Summary: The capacity for macrophages to polarize into distinct functional activation states is critical for tuning inflammatory responses. In addition to IL-4/IL-13, signals from the inflammatory milieu can further modulate M2 marker expression, with cAMP playing a role in increasing M2 markers through a PKA/C/EBP beta/CREB dependent pathway.

Critical considerations for the development of potency tests for therapeutic applications of mesenchymal stromal cell-derived small extracellular vesicles

Summary: Mesenchymal stromal/stem cells (MSCs) have been approved for treating certain diseases through paracrine secretion of small extracellular vesicles (sEVs). Despite the potential advantages of sEV preparations over cellular MSC products, there are challenges in developing MSC-sEV drug products, including establishing quality control metrics and developing effective potency assays.

PD-L1 induces macrophage polarization toward the M2 phenotype via Erk/Akt/mTOR

Yi Wei et al.

Summary: PD-L1 binds to PD-1 and promotes M2 polarization accompanied by mitochondrial function enhancement and metabolic reprogramming via Erk/Akt/mTOR. This study elucidates the role of PD-L1 in macrophage polarization and verifies the underlying mechanisms for the first time. Considering that aberrantly upregulated PD-L1 expression contributes to a wide variety of diseases, targeting PD-L1-mediated macrophage polarization is a prospective therapeutic strategy for both neoplastic and nonneoplastic diseases.

Microglial Adenosine Receptors: From Preconditioning to Modulating the M1/M2 Balance in Activated Cells

Rafael Franco et al.

Summary: Neuronal survival relies on glial support, with activated microglia showing either harmful pro-inflammatory (M1) or beneficial neuroprotective (M2) phenotypes; maintaining the M1/M2 balance is crucial in neuroinflammation. Adenosine receptors play a significant role in increasing M2 cell numbers and preconditioning for better resilience against damaging events. The potential therapeutic targeting of adenosine receptors and adenosine-cannabinoid complexes for microglia-mediated neuroprotection is discussed.

The Macrophage Response Is Driven by Mesenchymal Stem Cell-Mediated Metabolic Reprogramming

Noymar Luque-Campos et al.

Summary: MSCs, multipotent adult stromal cells, are widely studied for their regenerative and immunomodulatory properties, capable of modulating macrophage plasticity and function. Metabolic changes can also affect macrophage fate and function, prompting phenotype change in response to environmental cues. Understanding the crosstalk between macrophages and MSCs can improve therapeutic potential in inflammatory diseases, cancer, and tissue repair processes.
