Effect of PEG Anchor and Serum on Lipid Nanoparticles: Development of a Nanoparticles Tracking Method

Summary: Polyethylene glycol (PEG) is used to enhance the stealth properties of Lipid Nanoparticles (LNPs) and its anchoring to membranes impacts the fate of LNPs in vivo. The behavior of LNPs can be predicted based on the dissociation of lipid-PEG using Nanoparticle Tracking Analysis (NTA) in serum. Different lipid-PEG formulations were prepared and their interactions with serum components were monitored using NTA, which showed that the efficiency of LNPs varied depending on the lipid-PEG used. CSL3 lipid and Cer-PEG were found to be promising for LNP formulation.
