Novel smac mimetic ASTX660 (Tolinapant) and TNF-α synergistically induce necroptosis in bladder cancer cells in vitro upon apoptosis inhibition

Lei Wang et al.

Summary: In this study, a novel Smac mimetic, ASTX660, was shown to induce necroptosis in apoptosis-insensitive bladder cancer (BC) cells. Mechanistically, ASTX660 promoted the formation of the necrosome complex, and its efficacy was dependent on TNF-alpha signaling and IRF1 after caspase inhibition. These findings suggest that ASTX660 can overcome the limitation of apoptosis induction and provide important clues for the design of a novel treatment strategy for BC.

Hallmarks of Cancer: New Dimensions

Douglas Hanahan

Summary: The concept of the hallmarks of cancer is a tool for simplifying the complexity of cancer phenotypes and genotypes into basic principles. This article proposes phenotypic plasticity and disrupted differentiation as distinct hallmark capabilities, and discusses nonmutational epigenetic reprogramming and polymorphic microbiomes as enabling characteristics. Additionally, the importance of senescent cells in the tumor microenvironment is highlighted. The integrative concept of the hallmarks of cancer helps in understanding the mechanisms of cancer development and applying that knowledge to cancer medicine.

Urinary biomarkers in bladder cancer: A review of the current landscape and future directions

Kenrick Ng et al.

Summary: This narrative review discusses established and emerging urinary biomarkers for non-muscle invasive bladder cancer, highlighting the limitations of classical FDA-approved protein biomarkers. Advancements in epigenetic and genetic markers as well as extracellular vesicles have shown promise in enhancing sensitivity and specificity in bladder cancer diagnosis. The future focus will be on utilizing these markers in combination to create predictive signatures for everyday clinical practice.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Killing by Degradation: Regulation of Apoptosis by the Ubiquitin-Proteasome-System

Ruqaia Abbas et al.

Summary: Apoptosis is a crucial process for cell health and development, regulated by pro- and anti-apoptotic proteins with the ubiquitin proteasome system playing a key role in protein turnover and apoptosis modulation. Targeting these pathways presents opportunities for innovative anti-cancer therapies by recruiting the UPS for selective cancer cell killing.

Evaluation of hTERT, KRT7, and survivin in urine for noninvasive detection of bladder cancer using real-time PCR

Reza Yahyazadeh et al.

Summary: The detection of bladder cancer using hTERT, KRT7, and SVV in urine samples shows higher sensitivity compared to urine cytology, with a combined use of these markers showing a significant improvement in the diagnostic capabilities, especially for low-grade TCC. The sensitivity of these biomarkers is not correlated with the grades and stages of bladder tumors, providing a promising noninvasive diagnostic method for follow-up of low-grade TCC in the bladder.
