Transcriptomic taxonomy and neurogenic trajectories of adult human, macaque, and pig hippocampal and entorhinal cells

Daniel Franjic et al.

Summary: The hippocampal-entorhinal system supports cognitive functions and has neurogenic capabilities in many species. This study profiled the single-nucleus transcriptomes in different subregions of the hippocampal-entorhinal system in humans, macaques, and pigs. The findings revealed species differences in neurogenesis and cellular diversity, as well as the association between certain proteins and Alzheimer's disease.

A pan-cancer single-cell transcriptional atlas of tumor infiltrating myeloid cells

Sijin Cheng et al.

Summary: This study conducted a pan-cancer analysis of single myeloid cells from 210 patients across 15 human cancer types, revealing distinct features of TIMs in different cancer types. For example, mast cells in nasopharyngeal cancer were associated with better prognosis, and pro-angiogenic TAMs showed diverse markers across different cancer types. The composition of TIMs appeared to be linked with certain features of somatic mutations and gene expressions.

Comparative cellular analysis of motor cortex in human, marmoset and mouse

Trygve E. Bakken et al.

Summary: The research reveals a broadly conserved cellular makeup of the primary motor cortex in humans, marmoset monkeys, and mice, with species-specific specializations. Few cell-type marker genes are conserved across species, indicating some candidate genes and regulatory mechanisms responsible for functional identity of cell types and their species-specific adaptations.

Single-nucleus transcriptome analysis reveals cell-type-specific molecular signatures across reward circuitry in the human brain

Matthew N. Tran et al.

Summary: Single-cell gene expression technologies were used to study cell types across five key nodes of the human brain reward circuitry, identifying novel neuron subpopulations and characterizing specific inhibitory cell populations. The study also revealed unique patterns of transcriptomic dynamics and identified genetic risk associated with psychiatric disease and addiction in subpopulations of MSNs in the nucleus accumbens.

Neuregulin-1-dependent control of amygdala microcircuits is critical for fear extinction

Ming Chen et al.

Summary: The NRG1-ErbB4 pathway in the intercalated cells mass of amygdala (ITC) promotes fear extinction by increasing feed-forward inhibition of CeM neurons through enhanced GABAergic neurotransmission. The pathway may regulate fear extinction through P/Q-type voltage-activated Ca2+ channels in ITC. Overall, this pathway in ITC could be a potential target for treating anxiety disorders.

A cellular and spatial map of the choroid plexus across brain ventricles and ages

Neil Dani et al.

Summary: This study presents a single-cell and spatial atlas of each ChP in the mouse brain, highlighting diverse cell types, states, and expression programs in different ventricles at different stages of development, maturation, and aging. The atlas reveals regionalized expression patterns of epithelial and fibroblast cells by ventricle, with significant transcriptional shifts during maturation and minor shifts in aged cell types. Epithelial cells upregulate host-defense programs with aging, while resident macrophages upregulate IL-1 beta signaling genes.

Expression of FoxP2 in the basal ganglia regulates vocal motor sequences in the adult songbird

Lei Xiao et al.

Summary: The disruption of the transcription factor FoxP2 in the basal ganglia impacts vocal development in humans and songbirds. FoxP2 expression in the basal ganglia is essential for the fluent initiation and termination of birdsong, as well as the maintenance of song syllable sequencing. Knockdown of FoxP2 leads to imbalanced dopamine receptor expression and disrupts vocal motor sequencing, implicating dopaminergic signaling in regulating song production.

Intercalated amygdala clusters orchestrate a switch in fear state

Kenta M. Hagihara et al.

Summary: Our study revealed that distinct inhibitory clusters of intercalated neurons in the mouse amygdala play diametrically opposed roles in the acquisition and retrieval of fear extinction memory, antagonizing each other through mutual synaptic inhibition. The balance of activity between these clusters represents a unique regulatory motif that orchestrates a distributed neural circuitry regulating the switch between high- and low-fear states. These findings suggest a broader role for ITCs in various amygdala functions underlying the ability to adapt to significant environmental demands.

Midbrain dopaminergic inputs gate amygdala intercalated cell clusters by distinct and cooperative mechanisms in male mice

Ayla Aksoy-Aksel et al.

Summary: Dopaminergic signaling is crucial for associative learning, particularly in fear and extinction learning. Intercalated cell (ITC) clusters within the amygdala receive dense dopaminergic projections, with novel mechanisms by which dopamine controls ITCs discovered in mice. These findings shed light on the dopaminergic mechanisms that regulate the balance between different ITC clusters, impacting fear behavior.
