Modifications in the piperazine ring of nucleozin affect anti-influenza activity

Erick Correa-Padilla et al.

Summary: This study analyzed piperazine-modified nucleozin analogs to increase conformational freedom, contributing to the research of nucleozin analogs' discovery and design. A new synthetic strategy to obtain nucleozin and its analogues was also described, and two of them were evaluated in vitro. Although the analogues were less active, the loss of activity highlighted the significance of the piperazine ring for maintaining the activity of nucleozin analogs. This result agrees with the prediction of anti-influenza activity made with a QSAR model presented in this work. The proposed model and synthetic route will be useful for the further development of nucleozin analogs with antiviral activity.

Detection of Highly Pathogenic Avian Influenza Virus H5N1 Clade 2.3.4.4b in Great Skuas: A Species of Conservation Concern in Great Britain

Ashley C. Banyard et al.

Summary: The UK and Europe have experienced consecutive outbreaks of highly pathogenic avian influenza during the 2020/21 and 2021/22 autumn/winter seasons, resulting in significant mortalities among land- and waterfowl. Understanding the epidemiology and transmission of these viruses in different species is crucial for implementing effective control measures. Infections in different species can range from mild or asymptomatic to severe cases with high morbidity and mortality rates. Defining disease outcomes in different species is essential for understanding their role in the maintenance and transmission of these pathogens.

Global update on the susceptibilities of human influenza viruses to neuraminidase inhibitors and the cap-dependent endonuclease inhibitor baloxavir, 2018-2020

Summary: Global analysis showed low susceptibility of influenza viruses to neuraminidase inhibitors (NAIs) and the polymerase acidic (PA) inhibitor baloxavir. NA-H275Y substitution in A(H1N1)pdm09 viruses conferred high resistance to oseltamivir and peramivir. Baloxavir showed reduced susceptibility in a small percentage of viruses, with Japan having a higher rate. Overall, NAIs and baloxavir remain appropriate choices for treating influenza virus infections.

A multivalent nucleoside-modified mRNA vaccine against all known influenza virus subtypes

Claudia P. Arevalo et al.

Summary: It is important to develop a vaccine that can provide cross-reactive and subtype-specific antibodies as seasonal influenza vaccines offer limited protection against pandemic influenza virus strains. This study developed a multivalent vaccine using nucleoside-modified mRNA encoding hemagglutinin antigens from all known influenza virus subtypes, which elicited high levels of antibodies in mice and ferrets that reacted to multiple antigens. Vaccination with this mRNA vaccine protected mice and ferrets from both matched and mismatched viral strains.
