The Pathological Activation of Microglia Is Modulated by Sexually Dimorphic Pathways

Jennifer L. O'Connor et al.

Summary: Microglia, the primary immunocompetent cells in the CNS, play a critical role in maintaining CNS homeostasis and can exhibit a range of responses, from pro-inflammatory neurotoxic to anti-inflammatory protective, depending on their local cues. This review aims to define the developmental and environmental factors that influence microglial polarization and discusses the potential influence of sexually dimorphic factors. Furthermore, the review highlights the disparities in disease severity or diagnosis rates between males and females in various CNS disorders and proposes that microglial sexual dimorphism may underlie these differences.

Osteopontin promotes hepatocellular carcinoma progression through inducing JAK2/STAT3/NOX1-mediated ROS production

Qipeng Wu et al.

Summary: This study demonstrates that osteopontin (OPN) promotes hepatocellular carcinoma (HCC) progression by increasing reactive oxygen species (ROS) production through upregulating the expression of NADPH oxidase 1 (NOX1) and activating the JAK2/STAT3 signaling pathway. Disruption of the OPN/NOX1 axis might be a promising therapeutic strategy for HCC.

Deciphering Tumour Heterogeneity: From Tissue to Liquid Biopsy

Pauline Gilson et al.

Summary: Malignant tumors exhibit high levels of heterogeneity at both molecular and phenotypic levels, which poses challenges for patient management. This heterogeneity exists between patients and even within a single patient over space and time. Current treatment decision-making often only considers the molecular characteristics of a limited tumor tissue sample, without taking into account the complexity and continuous evolution of bulk tumors. This review explores the extent of tumor heterogeneity, focusing on intra-tumor heterogeneity (ITH), and discusses the mechanisms that promote and sustain this diversity in cancers. The clinical implications of ITH in the management of cancer patients are summarized, along with discussions on current approaches to adequately appreciate ITH.

Urine Cell-Free MicroRNAs in Localized Prostate Cancer Patients

Yoko Koh et al.

Summary: This study aimed to explore the potential of urine cell-free microRNAs (cfmiRs) as diagnostic biomarkers for early-stage prostate cancer (PCa). The results showed that specific cfmiRs in urine samples may have utility in the detection of PCa and could complement or improve currently available screening assays.

STAT3-EMT axis in tumors: Modulation of cancer metastasis, stemness and therapy response

Mehrdokht Sadrkhanloo et al.

Summary: Epithelial-to-mesenchymal transition (EMT) mechanism is responsible for tumor metastasis and inhibition of this process improves prognosis. STAT3 signaling is shown to promote EMT-mediated cancer migration and invasion, and its inhibition suppresses cancer metastasis and improves therapy response.

Site-specificmetastasis: A cooperation between cancer cells and the metastatic microenvironment

Sivan Izraely et al.

Summary: This review examines the collaboration between disseminating tumor cells (DTC) and the microenvironment of future metastatic organ sites in the establishment of organ-specific metastasis. It discusses targeted migration, transmigration, seeding, and colonization of DTC, as well as the impact of DTC-microenvironment interactions on tumor dormancy and response to therapy. Additionally, it summarizes studies focusing on the effects of the microenvironment on a specific site-specific metastasis, such as brain metastasis.

Charlotte M. Flynn et al.

Summary: The study highlights the significance of IL-6 receptors internalization in cell responsiveness and signaling activation, emphasizing the important role of IL-6R and gp130 internalization in signal cascade activation. The recycling of IL-6R and gp130 back to the cell surface after internalization, enhanced by IL-6, is also emphasized as crucial for signaling activation.

New Frontiers in Cancer Imaging and Therapy Based on Radiolabeled Fibroblast Activation Protein Inhibitors: A Rational Review and Current Progress

Surachet Imlimthan et al.

Summary: The tumor microenvironment (TME) has become a new paradigm of cancer diagnosis and therapy in the past decade, with cancer-associated fibroblasts (CAFs) being identified as critical stromal cells that regulate tumor growth and progression. Research on FAP-based radiopharmaceuticals for visualizing malignancies and delivering theranostic radiopharmaceuticals to the TME has been intensively investigated. This review provides an overview of TME compositions, particularly CAFs and FAP, and their roles in cancer biology, as well as the challenges and requirements for radiolabeled FAP inhibitors in clinical translation.

Summary: The study found that miR-200b was increased in the exosomes of ovarian cancer patients, promoting macrophage M2 polarization and enhancing cell viability and invasion of ovarian cancer cells. Additionally, miR-200b inhibited KLF6 expression, while overexpression of KLF6 rescued miR-200b-induced macrophage polarization. The results suggested that miR-200b might be a potential target for ovarian cancer treatment.
