Comparison of concentrated fresh mononuclear cells and cultured mesenchymal stem cells from bone marrow for bone regeneration

Summary: Autologous bone marrow mononuclear cell (BMMNC) transplantation has been studied extensively, showing that concentrated fresh BMMNCs may be superior to cultured bone marrow-derived mesenchymal stem cells (BMSCs) in promoting bone regeneration in segmental bone defects in Beagle dogs. The results revealed that the BMMNC group had significantly more bone regeneration with better mineralization and similar properties to native bone tissue.

Summary: Three-dimensional spheroid culture can promote osteogenic differentiation of bone marrow mesenchymal stromal cells, while 3D printing allows for customized scaffolds for complex bone defects. This study compared the potential of human BMSC cultured as 2D monolayers or 3D spheroids in constructs for bone regeneration. Results showed a trend for superior in vitro mineralization with 3D BMSC, but similar in vivo performance between 2D and 3D BMSC, indicating PLATMC-HPLG constructs as promising scaffolds for bone tissue engineering.

Real-time assessment of guided bone regeneration in critical size mandibular bone defects in rats using collagen membranes with adjunct fibroblast growth factor-2

Summary: This study investigated the bone healing pattern in rat mandibular bone defects with and without RCM containing FGF-2. Real-time assessment revealed that RCM/FGF-2 effectively promoted bone regeneration within the critical-size mandibular defects in rats, with increased volume, BMD, and closure percentage of the NFB area compared to control and RCM groups. Histological evaluation confirmed the presence of osteoblasts, osteocytes, and blood vessels within the newly formed bone.
