A Neuron, Microglia, and Astrocyte Triple Co-culture Model to Study Alzheimer's Disease

Celia Luchena et al.

Summary: Glial cells play an essential role in the progression of Alzheimer's disease. A straightforward and reproducible triple co-culture model of neurons, astrocytes, and microglia was developed to study AD. This model provides a better representation of physiological conditions compared to individual primary cultures and can be used to study neurodegeneration and neuroinflammation in the context of AD and other neurodegenerative diseases.

A primary rodent triculture model to investigate the role of glia-neuron crosstalk in regulation of neuronal activity

Leela Phadke et al.

Summary: Neuroinflammation and hyperexcitability play important roles in the pathogenesis of neurodegenerative diseases. To study the cellular interactions involved in these processes, a new triculture system containing neurons, astrocytes and microglia was developed. The presence of the intended cell types was confirmed and it was found that microglia suppressed neuronal activity in a dose-dependent manner in the triculture model. This functional triculture model provides a more relevant and complete platform for studying glia-neuron interactions and drug discovery.

Anti-inflammatory clearance of amyloid-β by a chimeric Gas6 fusion protein

Hyuncheol Jung et al.

Summary: In this study, an engineered protein was developed to induce the engulfment of amyloid-beta (Aβ), leading to improved behavior in Alzheimer's disease mouse models. This protein showed better outcomes compared to conventional antibody therapy, without causing increased inflammation or vascular pathology.

Influence of microchannel geometry on device performance and electrophysiological recording fidelity during long-term studies of connected neural populations

Noah Goshi et al.

Summary: The study investigates the impact of microchannel geometry and cell seeding density on the performance of compartmentalized microfluidic neural cell culture platforms. It concludes that microchannel height is the primary determinant of device performance, while other parameters offer additional flexibility for customization. This research provides design rules for engineering microfluidic neural culture platforms and demonstrates their utility in long-term studies.

Microglia as hackers of the matrix: sculpting synapses and the extracellular space

Joshua D. Crapser et al.

Summary: Microglia play a crucial role in shaping the synaptic environment in the brain, including modulating the perisynaptic matrix and perineuronal nets (PNNs). Alterations in the extracellular matrix (ECM) surrounding synapses may have implications for synapse dynamics, highlighting the interconnected relationship between microglia, synapses, and the ECM. Further research is needed to understand the mechanisms by which microglial regulation of the ECM impacts normal brain homeostasis and diseases.
