The MLL3/4 complexes and MiDAC co-regulate H4K20ac to control a specific gene expression program

Xiaokang Wang et al.

Summary: This study reveals the important role of MiDAC in embryonic development and neurite outgrowth, and its functional interaction with UTX and MLL3/4 complexes. MiDAC acts as a negative regulator of H4K20ac and inhibits the expression of associated genes at the transcriptional level.

Integrating quantitative proteomics with accurate genome profiling of transcription factors by greenCUT&RUN

Sheikh Nizamuddin et al.

Summary: The greenCUT&RUN method utilizes GFP-tagging and GFP-specific nanobody to profile transcription regulators with high sensitivity, resolution, accuracy, and reproducibility, while eliminating antibody dependency and variability. It allows for rapid profiling of mutant chromatin and transcription proteins, and is a versatile and widely applicable technique for genome mapping.

UTX condensation underlies its tumour-suppressive activity

Bi Shi et al.

Summary: UTX, a histone H3K27 demethylase, is an important tumour suppressor frequently mutated in human cancers. This study reveals that phase separation of UTX underlies its chromatin-regulatory activity in tumour suppression. UTX recruits histone methyltransferase MLL4 to condensates and enriches MLL4's activity, demonstrating the crucial role of liquid condensates in the tumour-suppressive activity of a chromatin regulator.
