Defining biomarkers in oral cancer according to smoking and drinking status

Juliette Rochefort et al.

Summary: This study found that smokers and drinkers have different immune profiles compared to non-smokers and non-drinkers in terms of cellular composition and cytokine expression in the tumor microenvironment, which may be useful for prognosis.

Oral squamous cell carcinoma arising from chronic traumatic ulcers

Gerardo Gilligan et al.

Summary: This study describes the clinicopathological features of non-healing chronic traumatic ulcers (NHCTU) that later developed into oral squamous cell carcinoma (OSCC). The study found that a subset of chronic traumatic ulcers may have the potential for malignant transformation.

Molecular drivers of oral cavity squamous cell carcinoma in non-smoking and non-drinking patients: what do we know so far?

Sophia Uddin et al.

Summary: OCSCC is one of the most common head and neck cancers worldwide, with risk factors including tobacco and excess alcohol consumption. However, in recent years, there has been an increase in the incidence of OCSCC in patients without these traditional risk factors. The cause of this increase is unclear, and it is believed that various genetic, environmental, and infectious factors may play a role. Additionally, oral cancer in non-smoking, non-drinking patients may have a distinct phenotype resulting in more aggressive disease presentation and poorer prognosis.

The role of tumor-associated macrophages in oral squamous cell carcinoma

Summary: This article reviews the role of tumor-associated macrophages (TAMs) in the invasion, metastasis, angiogenesis, and immunosuppression of oral squamous cell carcinoma (OSCC), as well as discusses the therapeutic strategies targeting TAMs for the treatment of OSCC.

Oral potentially malignant disorders: A consensus report from an international seminar on nomenclature and classification, convened by the WHO Collaborating Centre for Oral Cancer

Saman Warnakulasuriya et al.

Summary: Oral potentially malignant disorders (OPMDs) are associated with an increased risk of oral cancer, and an expert group updated the nomenclature and classification of OPMDs, adding oral lichenoid lesions and oral manifestations of 'chronic graft-versus-host disease' to the list. It is recommended to establish a global research consortium to study the natural history of OPMDs based on the proposed classification.

Genomic Signature of Oral Squamous Cell Carcinomas from Non-Smoking Non-Drinking Patients

Kendrick Koo et al.

Summary: A distinct cohort of non-smoking non-drinking patients with oral cavity squamous cell carcinomas have been identified, showing specific genomic profiles different from patients who smoke and/or drink, with associations to mutations in TP53, CDKN2A, EGFR, and BRCA2 genes. Further exploration is needed to elucidate the molecular aetiology in these patients.

Impact of smoking on dendritic cells in patients with oral squamous cell carcinoma

Heron Stahelin et al.

Summary: Smoking is associated with a reduction in dendritic cell density in oral squamous cell carcinoma patients, especially in the surrounding areas. Poorly differentiated tumors have lower density of immature DCs in the peritumoral region.

Tumor-infiltrating lymphocytes in the immunotherapy era

Sterre T. Paijens et al.

Summary: The success of cancer immune checkpoint blockade has led to increased interest in tumor-infiltrating lymphocytes (TILs) across different cancer types. Single-cell analysis has revealed high cellular heterogeneity in TILs, with distinct patterns of immune activation and exhaustion. While TIL distributions are similar across tumor types, their responses to immunotherapy vary, highlighting the complexity of tumor-immune interactions.
