Sirt6 protects cardiomyocytes against doxorubicin-induced cardiotoxicity by inhibiting P53/Fas-dependent cell death and augmenting endogenous antioxidant defense mechanisms

Sisi Wu et al.

Summary: Sirt6 plays a crucial role in mitigating oxidative stress and cardiac injury by upregulating endogenous antioxidants and suppressing ROS-stirred p53-dependent oxidative stress and cell death signaling pathways, thereby protecting the heart against DOX-induced cardiotoxicity.

Mechanism of allosteric activation of SIRT6 revealed by the action of rationally designed activators

Shaoyong Lu et al.

Summary: The discovery of activator compounds binding to an allosteric site on SIRT6 has potential implications for aging-related and cancer diseases. The mechanism of allosteric activation by MDL-801 involves conformational rotation of the substituent on the R-ring, leading to increased deacetylation activity. The presence of the 5-Br atom in MDL-801 may serve as an allosteric driver controlling the ligand's effectiveness.

Prohibitin depletion extends lifespan of a TORC2/SGK-1 mutant through autophagy and the mitochondrial UPR

Patricia de la Cruz-Ruiz et al.

Summary: Mitochondrial prohibitins (PHB) play a role in regulating lifespan in animals, affecting various metabolic pathways. Depletion of PHB extends the lifespan of metabolically compromised mutants like sgk-1, while impacting mitochondrial homeostasis and lipid metabolism. The longevity effect of PHB depletion in sgk-1 mutants is associated with the induction of UPRmt and autophagy, but not mitophagy, suggesting a complex interplay of pathways contributing to increased lifespan.

Cardiac Energy Metabolism in Heart Failure

Gary D. Lopaschuk et al.

Summary: Changes in cardiac energy metabolism are complex and depend on the severity and type of heart failure, as well as the presence of common comorbidities such as obesity and type 2 diabetes. The failing heart experiences an energy deficit due to decreased mitochondrial oxidative capacity, compensated for by increased ATP production from glycolysis. Alterations in mitochondrial ATP production fuels and fatty acid oxidation contribute to the inefficiency of the failing heart, with potential therapeutic targets in energy metabolic pathways to improve cardiac function.

A novel SIRT6 activator ameliorates neuroinflammation and ischemic brain injury via EZH2/FOXC1 axis

Tailin He et al.

Summary: Ischemic stroke is a major cause of death globally, and controlling neuroinflammation is a challenge. The novel SIRT6 activator MDL-811 inhibits inflammation by regulating the Foxc1 gene and promoting deacetylation of EZH2, showing potential therapeutic effects in ischemic stroke.

Association of SIRT6 circulating levels with urinary and glycometabolic markers in pre-diabetes and diabetes

Che Bian et al.

Summary: The study aimed to detect the expression of serum Sirtuin 6 (SIRT6) and urinary albumin creatinine ratios (UACR) in type 2 diabetes mellitus (T2DM) patients. Results showed that with the aggravation of diabetic status, HbA1c, TNF alpha, HIF1 alpha, and SIRT6 increased significantly. Correlation analysis demonstrated that SIRT6 was significantly positively correlated with glycolipid metabolism and correlated with UACR, TNF alpha, and HIF1 alpha in T2DM patients. Ridge regression analysis showed that SIRT6 was a risk factor for both glycolipid metabolism and urinary protein.

Intermittent fasting in the prevention and treatment of cancer

Katherine K. Clifton et al.

Summary: Intermittent fasting (IF) has gained popularity as a potential anticarcinogenic strategy, but its effects on cancer patients remain unclear without high-quality randomized clinical trials. More research is needed to determine the risks and benefits of IF for cancer patients before it can be recommended outside of clinical trials.

SIRT6 transcriptionally regulates fatty acid transport by suppressing PPARγ

Danish Khan et al.

Summary: Pathological lipid accumulation in cardiomyocytes is associated with enhanced fatty acid uptake through specific transporters, regulated by the transcription factor SIRT6. SIRT6 deficiency leads to increased expression of fatty acid transporters and lipid accumulation, with its interaction with PPARγ being critical in regulating transporter expression.്
