Garlic (Allium sativum L.): Its Chemistry, Nutritional Composition, Toxicity, and Anticancer Properties

Abdur Rauf et al.

Summary: The current review discusses the chemistry, nutritional composition, toxicity, and biological functions of garlic and its bioactive compounds against various types of cancers. The study finds that garlic contains several phytoconstituents with activities against cancer, and these compounds exhibit mechanistic actions that can inhibit the induction, development, and progression of cancer. Moreover, garlic compounds also regulate multiple signaling cascades and have the potential for developing as antitumor drugs.

Diallyl disulfide and diallyl trisulfide in garlic as novel therapeutic agents to overcome drug resistance in breast cancer

RamaRao Malla et al.

Summary: Breast cancer is a major cause of cancer-related deaths in women worldwide. The increase in tumor heterogeneity and aggressiveness has made the treatment of breast cancer challenging. High intake of certain Allium vegetables, such as garlic, has been associated with a reduced risk of developing breast cancer. DADS and DATS, compounds found in garlic, have anticancer activity and can interfere with breast cancer cell proliferation and metastasis. This review focuses on multidrug resistance mechanisms in breast cancer and discusses the potential anticancer activities of DADS and DATS, particularly in triple-negative breast cancer.

Epigenetic modulation of immunotherapy cofactors to enhance tumor response in lung cancer

Anuhya Kommalapati et al.

Summary: Immunotherapy with checkpoint inhibitors has replaced chemotherapy in some settings, allowing many patients to live longer with fewer side effects, but only works for about one-fifth of unselected patients. Important cofactors within the tumor microenvironment, such as T-cells and chemokines, can impact the efficacy of immunotherapy through epigenetic modulation.

Two-stage Study of Familial Prostate Cancer by Whole-exome Sequencing and Custom Capture Identifies 10 Novel Genes Associated with the Risk of Prostate Cancer

Daniel J. Schaid et al.

Summary: This study uses a two-stage design to identify new genetic variants associated with prostate cancer (PCa) in individuals with a family history of the disease or with a more aggressive form of PCa. The research detected 11 known genes associated with PCa and 10 novel genes, most of which are primarily linked to aggressive PCa risk.

Nongenetic Evolution Drives Lung Adenocarcinoma Spatial Heterogeneity and Progression

Daniele Tavernari et al.

Summary: This study provides a detailed molecular map of intratumor spatial heterogeneity in lung adenocarcinoma, revealing non-genetic routes of cancer evolution. The transition from indolent to aggressive patterns in tumors is driven by epigenetic and transcriptional reprogramming reshaping cancer cell identity rather than genetic alterations. Highly multiplexed protein spatial profiling shows the coexistence of immune desert, inflamed, and excluded regions within individual tumors, matching histologic pattern composition.

Nuclear mechanisms of gene expression control: pre-mRNA splicing as a life or death decision

Jackson M. Gordon et al.

Summary: This article discusses the role of retained intron RNAs in shaping gene expression programs and the regulation of the biogenesis and fate of RI-RNAs by nuclear organization. Proximity of RNA to nuclear speckles may affect choices ranging from efficient co-transcriptional splicing, export and stability to regulated post-transcriptional splicing and possible vulnerability to decay.

UHRF1 regulates alternative splicing by interacting with splicing factors and U snRNAs in a H3R2me involved manner

Peng Xu et al.

Summary: The well-established functions of UHRF1 in DNA biological processes have been expanded to show that UHRF1 also plays a role in RNA metabolism by regulating alternative RNA splicing. The interaction of UHRF1 with various splicing factors and its involvement in U snRNAs suggest a complex mechanism at play, providing insights into UHRF1-mediated alternative RNA splicing and its implications in disease development.

NucHMM: a method for quantitative modeling of nucleosome organization identifying functional nucleosome states distinctly associated with splicing potentiality

Kun Fang et al.

Summary: A novel computational method, NucHMM, was developed to identify functional nucleosome states associated with cell type-specific combinatorial histone marks and nucleosome organization features. Testing on public data from MCF7, H1, and IMR90 cells revealed 11 distinct functional nucleosome states, which were shown to be significantly associated with splicing potentiality. This study advances our understanding of chromatin function at the nucleosome level and provides insights into the interplay between nucleosome organization and splicing processes.

Genetic and non-genetic clonal diversity in cancer evolution

James R. M. Black et al.

Summary: This article discusses the importance of functional and non-functional intra-tumour heterogeneity (ITH) in cancer evolution. It emphasizes the need to consider genetic and non-genetic factors in tumor evolution, and highlights the role of systems-biology analytical approaches in understanding the functional ITH. Ultimately, deepening our understanding of tumour evolution through functional ITH will lead to the development of novel therapies and improved outcomes for patients.
