Kynurenine Pathway in Diabetes Mellitus-Novel Pharmacological Target?

Kamila Koziel et al.

Summary: The tryptophan-kynurenine pathway is important for tryptophan conversion in the brain and periphery, and kynurenines have diverse biological actions. Imbalance of beneficial and harmful kynurenines is implicated in the pathogenesis of neurodegenerative and metabolic disorders including diabetes mellitus. Inflammation affects the function of the pathway, and kynurenines can modulate the immune response. This review highlights the role of the Trp-KYN pathway in diabetes and insulin resistance, suggesting it as a potential target for preventive and therapeutic interventions.

Seizure modulation by the gut microbiota and tryptophan-kynurenine metabolism in an animal model of infantile spasms

Chunlong Mu et al.

Summary: The study found that the combination of antibiotics and a ketogenic diet can reduce the occurrence of infantile spasms and improve the efficacy of the ketogenic diet. Examination of the gut microbiota and metabolomics showed downregulation of indoleamine 2,3-dioxygenase I and upregulation of hippocampal kynurenic acid, a metabolite with antiepileptic effects. Inhibitors of indoleamine 2,3-dioxygenase I and antibiotics that inhibit kynurenine formation were effective in reducing spasms and elevating hippocampal kynurenic acid. Additionally, a fecal microbiota transplant experiment demonstrated the important role of the gut microbiota in improving spasms.

Development of a translational inflammation panel for the quantification of cerebrospinal fluid Pterin, Tryptophan-Kynurenine and Nitric oxide pathway metabolites

Jingya Yan et al.

Summary: The development of a targeted translational inflammation panel using LC-MS/MS has shown promise in providing early diagnosis, rapid treatment, and insights into neuroinflammatory mechanisms for diseases such as encephalitis and multiple sclerosis. The method exhibited good sensitivity, linearity, and accuracy, with potential for further study in larger groups of neurological diseases.

Mitochondrial Impairment: A Common Motif in Neuropsychiatric Presentation? The Link to the Tryptophan-Kynurenine Metabolic System

Masaru Tanaka et al.

Summary: The cytoplasmic inheritance of human chloramphenicol resistance, which is mediated by mitochondrial DNA (mtDNA), has been discovered for almost half a century. Mutations in mtDNA have been found to cause severe inheritable metabolic diseases and neurodegenerative diseases. Recent studies have shown that impaired mitochondrial functions and loss of mitochondrial stress resilience can influence animal behaviors. In addition, the activation of the tryptophan-kynurenine metabolic system, which is related to stress and inflammation, has been linked to the development of neurological and psychiatric disorders.

The type I interferonopathies: 10 years on

Yanick J. Crow et al.

Summary: The term 'type I interferonopathy' was coined 10 years ago to describe rare genetic diseases caused by aberrant upregulation of type I interferon signaling. Viral nucleic acid detection is crucial for effective immune response, but failure in self versus non-self discrimination can lead to Mendelian inborn errors of immunity characterized by upregulation of type I interferon signaling.

Jingya Yan et al.

Summary: The study aimed to explore cerebrospinal fluid metabolite features in acute neuroinflammatory diseases and identified potential biomarkers in the tryptophan-kynurenine pathway and nitric oxide pathway that strongly predict neuroinflammation. These alterations represent a useful potential panel for neuroinflammation and hold potential for clinical translation practice.

Bacterial Meningitis in the Absence of Pleocytosis in Children A Systematic Review

Petra Zimmermann et al.

Summary: Bacterial meningitis can present without pleocytosis, especially in children who experience symptoms shortly after onset. It is important to culture cerebrospinal fluid samples from patients with relatively short symptom duration, even if cell counts and biochemistry appear normal. A second lumbar puncture may be necessary in suspected cases of bacterial meningitis with initially normal CSF findings.

Proteomics reveals distinct mechanisms regulating the release of cytokines and alarmins during pyroptosis

Kshiti Phulphagar et al.

Summary: Macrophages release a variety of proteins during pyroptosis, including cytokines, alarmins, and lysosomal-cargo proteins. While most proteins are released due to cell lysis, some low-molecular-weight proteins can exit cells without cell lysis. Cytokines and alarmins are released through an ER-Golgi-dependent pathway as free proteins rather than through extracellular vesicles.

From ELISA to Immunosorbent Tandem Mass Spectrometry Proteoform Analysis: The Example of CXCL8/Interleukin-8

Mieke Metzemaekers et al.

Summary: This study introduces a novel methodology (ISTAMPA) that combines immunoaffinity sample preparation with nano-scale liquid chromatography-tandem mass spectrometry for proteoform analysis. The research demonstrates the presence of different NH2-terminal proteoforms of CXCL8 due to site-specific proteolytic modification, with specific proteoforms showing enhanced activity. An immunosorbent ion trap top-down mass spectrometry-based approach allows for simultaneous detection and quantification of full-length CXCL8(1-77), elongated CXCL8(-2-77) and all naturally occurring truncated CXCL8 forms in biological samples.
