Summary: This study reveals the crucial role of mitochondrial transcription factor A (TFAM) in the formation and function of alveolar macrophages (AMs). TFAM deficiency leads to reduced numbers and impaired maturation of AMs. TFAM regulates the maintenance of the AM compartment by enhancing AM proliferation and self-renewal while suppressing inflammatory gene expression. Additionally, influenza virus infection impairs TFAM expression and mitochondrial function in AMs.
