Clinical presentation and analysis of genotype-phenotype correlations in patients with malignant infantile osteopetrosis

Ehud Even-Or et al.

Summary: Malignant infantile osteopetrosis (MIOP) is a severe form of osteopetrosis with autosomal recessive inheritance. Early diagnosis is crucial for proper management of the disease, with common features including growth retardation, visual impairment, and cytopenias. High awareness of presenting signs is essential for prompt diagnosis of MIOP.

A Mild Case of Autosomal Recessive Osteopetrosis Masquerading as the Dominant Form Involving Homozygous Deep Intronic Variations in the CLCN7 Gene

Jochen G. Hofstaetter et al.

Summary: Osteopetrosis is a rare hereditary disease characterized by increased bone mass of poor quality. We report a case of adult male with classical clinical features of osteopetrosis, which genetic analysis revealed a possible mild form of autosomal recessive osteopetrosis.

Summary: Autosomal recessive osteopetrosis, also known as infantile malignant osteopetrosis (IMO), is a rare and serious disease that is often challenging to diagnose and can be fatal if left untreated. This case report describes an infant girl who was diagnosed with IMO and had a novel mutation in the CLCN7 gene.

Pathobiologic Mechanisms of Neurodegeneration in Osteopetrosis Derived From Structural and Functional Analysis of 14 ClC-7 Mutants

Eleonora Di Zanni et al.

Summary: ClC-7 is a chloride-proton antiporter that plays a critical role in various cellular functions, particularly in lysosomes and osteoclasts. Functional evaluation of ClC-7 mutations can provide insights into their pathogenicity, although challenges exist due to the protein's cellular localization. This study suggests a correlation between the lack of ClC-7/Ostm1 in lysosomes and severe neurodegeneration, shedding light on potential mechanisms underlying certain diseases.

Interaction of prenatal bisphenols, maternal nutrients, and toxic metal exposures on neurodevelopment of 2-year-olds in the APrON cohort

Jiaying Liu et al.

Summary: BPA and BPS exposure during pregnancy may affect children's neurodevelopment, with maternal nutrient status and neurotoxicant metal co-exposures playing important roles. Selenium and cadmium were significant predictors of neurodevelopment outcomes, and maternal selenium may protect against the adverse effects of bisphenols. Maternal cadmium was associated with poorer motor development, while not modifying the effects of bisphenols on motor development. Further research is needed to explore sex-specific effects of BPA and BPS on child development.

Transcriptomic and bioinformatic analysis of Clcn7-dependent Autosomal Dominant Osteopetrosis type 2. Preclinical and clinical implications

Iona Norwood et al.

Summary: Autosomal Dominant Osteopetrosis type 2 (ADO2) is a rare genetic disease affecting not only bones but also multiple organs. RNA deep sequencing analysis revealed alterations in gene ontology terms and pathways associated with bone metabolism and osteoclast biology. Additionally, enrichment of pathways related to tissue fibrosis and behavioral alterations were identified in soft organs affected by ADO2.

Benchmarking deep learning splice prediction tools using functional splice assays

Tabea V. Riepe et al.

Summary: Hereditary disorders are often caused by genetic variants affecting RNA splicing, but predicting the pathogenicity of variants in noncanonical splice sites and deep intronic regions is challenging. A study comparing splice prediction tools on variants in two genes found that SpliceRover and SpliceAI performed the best.

Efficient generation of osteoclasts from human induced pluripotent stem cells and functional investigations of lethal CLCN7-related osteopetrosis

Uta Roessler et al.

Summary: Human induced pluripotent stem cells (hiPSCs) have great potential for modeling human diseases and developing innovative therapeutic approaches. A novel differentiation method for forming functional osteoclasts from hiPSCs was reported, showing continuous production of monocyte-like cells for up to 9 weeks and generating sufficient material for multiple osteoclast differentiations. The study demonstrated that osteoclasts differentiated from hiPSCs could be used as a disease model for osteopetrosis and potentially other osteoclast-related diseases.

Clinical and molecular characterization of five Chinese patients with autosomal recessive osteopetrosis

Huanhuan Liang et al.

Summary: In this study, five Chinese children with osteopetrosis were identified with compound heterozygous variants of the CLCN7 and TCIRG1 genes through whole-exome sequencing. Three of these patients with TCIRG1 variants underwent hematopoietic stem cell transplantation, expanding the understanding of the relationship between genotype and clinical characteristics in osteopetrosis. The results deepen our knowledge of the gene variations associated with osteopetrosis.
