Risk Factors for Severe COVID-19 in Children: A Systematic Review and Meta-Analysis

Jae Hong Choi et al.

Summary: This study aimed to identify risk factors for severe COVID-19 in children and provide guidance for hospital admission and vaccination priority. The study found that comorbidities such as obesity, diabetes, heart disease, chronic lung diseases, neonates, and premature infants were associated with a higher prevalence of severe COVID-19 in children.

Multi-ancestry fine mapping implicates OAS1 splicing in risk of severe COVID-19

Jennifer E. Huffman et al.

Summary: The OAS1/2/3 cluster has been identified as a risk locus for severe COVID-19 among individuals of European ancestry, with a protective haplotype derived from Neanderthals. The splice variant of OAS1 is found in individuals of African ancestry independently, and it is likely responsible for the association with COVID-19 severity.

COVID-19 in Children: Where do we Stand?

Georgia B. Nikolopoulou et al.

Summary: From the beginning of the COVID-19 pandemic, it has been observed that children infected with SARS-CoV-2 are mostly asymptomatic or mildly symptomatic. The true prevalence of asymptomatic cases in children is likely underestimated and serologic surveys indicate that half of infected children show no symptoms. Children with COVID-19 have lower risk of hospitalization and severe complications.

The human genetic epidemiology of COVID-19

Mari E. K. Niemi et al.

Summary: In this article, the authors discuss how large-scale genomics studies are helping us to better understand the impact of host genetics on COVID-19 susceptibility and severity. These studies have identified genomic regions associated with the disease and provided insights into its pathology and potential therapeutic opportunities. The findings have implications for genetic risk prediction, drug repurposing, and understanding the long-term consequences of SARS-CoV-2 infection.

Detailed stratified GWAS analysis for severe COVID-19 in four European populations

Frauke Degenhardt et al.

Summary: Through in-depth genetic association analysis of COVID-19 patients and population controls, this study has identified genetic loci associated with severe COVID-19 and explored the potential effects of these loci. One association at 17q21.31 was found to be related to an inversion polymorphism creating two highly differentiated haplotypes. Additionally, a new locus at 19q13.33 was discovered, including the NAPSA gene which is primarily expressed in alveolar cells responsible for gas exchange in the lung.

A systematic review and meta-analysis of children with coronavirus disease 2019 (COVID-19)

Xiaojian Cui et al.

Summary: This study provides a comprehensive analysis of demographic characteristics, clinical symptoms, laboratory findings, and imaging features of COVID-19 in pediatric patients. It revealed that children with COVID-19 tend to have mild illness with typical manifestations of fever and cough, normal white blood cell counts, and common imaging features of normal or ground-glass opacity. However, the high incidence of critical cases and vomiting symptoms in children under 1 year old should be given attention.

Diagnosis of SARS-CoV-2 in children: accuracy of nasopharyngeal swab compared to nasopharyngeal aspirate

Giada Maria Di Pietro et al.

Summary: The study aimed to evaluate the diagnostic performances of nasopharyngeal swabs (NS) referred to nasopharyngeal aspirates (NPA) for SARS-CoV-2 in children. The results showed that NS has low sensitivity but high specificity in detecting SARS-CoV-2 in children.

Genetic mechanisms of critical illness in COVID-19

Erola Pairo-Castineira et al.

Summary: In critically ill patients with COVID-19, host genetic variants associated with antiviral defense mechanisms and inflammatory organ damage were identified through a genome-wide association study. These genetic signals provide potential targets for therapeutic development and repurposing of existing drugs. Large-scale randomized clinical trials will be essential to confirm the effectiveness of targeted treatment strategies.

Clinical characteristics, treatment and outcomes of paediatric COVID-19: a systematic review and meta-analysis

Omar Irfan et al.

Summary: This systematic review and meta-analysis compared paediatric COVID-19 disease characteristics, management, and outcomes based on World Bank country income level and disease severity. The majority of paediatric cases with COVID-19 recovered, but low- and middle-income countries had higher death rates among hospitalized children and lower ICU admission rates compared to high-income countries. Children with severe disease received more frequent antimicrobials, inotropes, and anti-inflammatory agents. Further global research on severe disease characterization in children with COVID-19 is needed.

Summary: This study aimed to analyze the spectrum of co-infections in pediatric COVID-19 patients and determine clinical and laboratory parameters predicting co-infection. Results showed that children with co-infections had higher morbidity and need for supportive therapy. Predictors of co-infection included localization of infection, neutrophil count, CRP levels, and hyperferritinemia.

Summary: The study suggests that HLA antigens may influence SARS-CoV-2 infection and the clinical evolution of COVID-19, while individuals with blood group A are at a greater risk of infection. These findings provide insights into the spread of the disease and may offer indications for infection prognosis and vaccination strategies.
