Characterization of a novel non-steroidal glucocorticoid receptor agonist optimized for topical treatment

Summary: This study describes a selective non-steroidal glucocorticoid receptor (GR) agonist, LEO 134310, for topical use, which is rapidly deactivated in the blood resulting in low systemic exposure and a higher therapeutic index compared with currently used glucocorticoids. The selectivity of LEO 134310 for GR was confirmed within a panel of nuclear receptors, and topical treatment with LEO 134310 in minipigs did not result in significant epidermal thinning.

The results showed acceptable reproducibility of telemetric measurement over a ten-week period in the pigs, but there were discrepancies between direct and indirect blood pressure measurements and the anesthesia protocols influenced blood pressure and heart rate.

Assessment of Two Commonly used Dermal Regeneration Templates in a Swine Model without Skin Grafting

Summary: Autologous skin grafting is still the gold standard for dermal replacement in medical care. However, modern wound dressings offer promising approaches to surface reconstruction. The behavior of two established dermal substitutes, a crosslinked and a non-crosslinked collagen biomatrix, was investigated in this study. It was found that both treatment groups demonstrated faster reepithelialization compared to the controls.
