Genetic correlates of phenotypic heterogeneity in autism

Varun Warrier et al.

Summary: Comprehensive factor analysis of core diagnostic features provides insights into the complex genetic architecture underlying phenotypic heterogeneity in autism. The study found that common genetic variants were associated with the core factors of autism, but not de novo variants. Higher autism polygenic scores were associated with a lower likelihood of co-occurring developmental disabilities, and autistic females showed a stronger over-inheritance of autism PGS compared to males.

A phenotypic spectrum of autism is attributable to the combined effects of rare variants, polygenic risk and sex

Danny Antaki et al.

Summary: This study reveals that the genetic etiology of autism spectrum disorder (ASD) is multifactorial, with rare genetic loads and polygenic risk associated with symptom severity. Additionally, parental age effects on offspring's risk for ASD are influenced by various genetic mechanisms.

Connectivity-Based Brain Network Supports Restricted and Repetitive Behaviors in Autism Spectrum Disorder Across Development

Anyi Zhang et al.

Summary: The study found improvements in restricted and repetitive behaviors with age in individuals with ASD, especially in stereotyped behaviors, ritualistic behaviors, and restricted behaviors. Functional connectivity analysis revealed that certain functional connections were associated with age and might contribute to these improvements.

Incidence, prevalence, and global burden of autism spectrum disorder from 1990 to 2019 across 204 countries

Marco Solmi et al.

Summary: This study analyzed the prevalence, incidence, and disability burden of autism spectrum disorder (ASD) worldwide using data from the Global Burden of Diseases, Injuries, and Risk Factors Study. The overall age-standardized estimates of ASD remained stable globally from 1990 to 2019, but the prevalence and disability-adjusted life years (DALYs) increased in high socio-demographic index (SDI) countries. In contrast, the age-standardized incidence decreased in some low SDI countries, indicating a need for improved awareness. The male/female ratio decreased over time, possibly due to increased attention to ASD in females.

Can the female protective effectliability threshold model explain sex differences in autism disorder?

Summary: Male sex is a strong risk factor for autism spectrum disorder (ASD), but the theory of a female protective effect (FPE) is not supported by current data.

Placental methylome reveals a 22q13.33 brain regulatory gene locus associated with autism

Yihui Zhu et al.

Summary: This study identified a novel ASD risk gene, NHIP, through whole genome bisulfite sequencing in placenta samples. NHIP is highly expressed in the brain and overlaps significantly with known ASD risk genes and NHIP-associated genes in the ASD brain. Additionally, a common structural variant associated with ASD risk, NHIP expression, and methylation levels was found.

Elevated Polygenic Burden for Autism Spectrum Disorder Is Associated With the Broad Autism Phenotype in Mothers of Individuals With Autism Spectrum Disorder

Kritika Nayar et al.

Summary: The study found associations between maternal pragmatic language and the child's social communicative atypicalities, as well as paternal rigid personality and increased repetitive behaviors in children. Maternal (but not paternal) ASD-PGSs were related to BAP traits, highlighting a potential female protective factor and implicating the polygenic etiology of ASD-related phenotypes in the BAP.

Individuals with common diseases but with a low polygenic risk score could be prioritized for rare variant screening

Tianyuan Lu et al.

Summary: Rare pathogenic variants are more prevalent among affected patients with a low PRS. Therefore, prioritizing individuals with disease but low PRS for sequencing may increase the yield of sequencing studies to identify rare variant heterozygotes.
