A near-complete genome assembly of Brassica rapa provides new insights into the evolution of centromeres

Summary: We have successfully completed a near-complete genome assembly of B. rapa Chiifu v4.0 using Oxford Nanopore Technology (ONT) ultralong-read sequencing and Hi-C technologies. This new assembly contains 12 contigs, with 8 chromosomes entirely reconstructed in a single contig. We found that the centromeres were mainly invaded by ALE and CRM long terminal repeats (LTRs), and there is a high divergence of centromere length and sequence among B. rapa genomes. This study provides valuable tools for genomic and genetic studies of Brassica species and sheds new light on the evolution of centromeres.

Assembly of the 81.6 Mb centromere of pea chromosome 6 elucidates the structure and evolution of metapolycentric chromosomes

Summary: In this study, the assembly and analysis of the pea metapolycentromere, a unique type of centromere that represents an intermediate stage between monocentric and holocentric organizations, is reported. The results reveal the sequence organization and potential mechanisms for the formation of the metapolycentromere. It is found that the metapolycentromere is formed through the expansion of centromeric chromatin and the accumulation of novel satellite repeats. Furthermore, genetic experiments demonstrate that centromere evolution involving chromosomal translocations and centromere repositioning also contribute to the formation of metapolycentromeres.

Gap-free genome assembly and comparative analysis reveal the evolution and anthocyanin accumulation mechanism of Rhodomyrtus tomentosa

Summary: This study presented a chromosome-level, gap-free T2T genome assembly of Rhodomyrtus tomentosa using PacBio and ONT long read sequencing. The assembled genome size was 470.35 Mb with 11 pseudochromosomes, and it contained 33,382 genes and 239.31 Mb of repetitive sequences. Phylogenetic analysis revealed the independent evolution of R. tomentosa and a recent whole-genome duplication event. The study also identified the major compounds of anthocyanins and their synthetic pathways in R. tomentosa and suggested that the coloring and high anthocyanin accumulation in R. tomentosa were determined by the activation of anthocyanin synthesis pathway.

Prospects of telomere-to-telomere assembly in barley: Analysis of sequence gaps in the MorexV3 reference genome
