Endogenous circadian reporters reveal functional differences of PERIOD paralogs and the significance of PERIOD:CK 1 stable interaction

Jiyoung Park et al.

Summary: Adverse consequences of a faulty circadian clock include sleep quality problems, poor short-term performance, and long-term risks of metabolic diseases and cancer. To study circadian disorders, researchers developed PER1-luc and PER2-luc circadian reporters in a validated cell model. Knocking out major clock genes in these cells revealed similarities with corresponding mutant mice, validating the platform for genetics studies. Differences between PER1 and PER2 were discovered, suggesting their unique roles in circadian rhythms and phosphorylation kinetics.

Molecular mechanisms of transgenerational epigenetic inheritance

Maximilian H. Fitz-James et al.

Summary: Increasing evidence suggests that non-DNA sequence-based epigenetic information can be inherited across generations in various organisms. Recent research has uncovered the signals and mechanisms underlying these epimutations, which vary among species and have a limited effect in mammals. However, common principles are emerging, with transmission occurring through direct replication or indirect reconstruction of the signals. This research improves our understanding of the contribution of DNA sequence and epigenetic variation to heritable phenotypic differences.

Maternal inheritance of glucose intolerance via oocyte TET3 insufficiency

Bin Chen et al.

Summary: Pregestational hyperglycemia increases the offspring's vulnerability to glucose intolerance. The insufficient demethylation by oocyte TET3 leads to hypermethylation in the paternal alleles of several insulin secretion genes. There is an environment-sensitive window in oocyte development that confers predisposition to glucose intolerance in the next generation.

Sperm-inherited H3K27me3 epialleles are transmitted transgenerationally in cis

Kiyomi Raye Kaneshiro et al.

Summary: The transmission of chromatin states from parent cells to daughter cells is important for preserving cell identity. A study on Caenorhabditis elegans showed that alleles inherited without a specific chromatin mark were more likely to be up-regulated in offspring cells, and the tissue context played a role in determining which genes were up-regulated. This study also demonstrated the transgenerational epigenetic transmission of chromatin states in C. elegans.

Circadian Misalignment and Metabolic Disorders: A Story of Twisted Clocks

Aurore Woller et al.

Summary: Mammalian physiological processes are regulated by a complex circadian clock network, which can adapt to changes in environmental conditions. Disruptions such as mistimed feeding or jet lag can lead to dysregulation of the genetic clockwork, impacting metabolic regulation and potentially causing diseases. Computational modeling can provide insights into this intricate system.

Sperm epigenetic alterations contribute to inter-and transgenerational effects of paternal exposure to long-term psychological stress via evading offspring embryonic reprogramming

Xiaoguo Zheng et al.

Summary: Research has shown that paternal exposure to long-term psychological stress can impact offspring through epigenetic alterations in sperm, specifically Differential DNA Methylation Regions (DMRs) related to stress response genes. These DMRs are inherited inter- and transgenerationally, passing paternal disorders epigenetically to offspring and evading embryonic reprogramming. Stress-induced dysregulation of sperm small non-coding RNAs may play a role in the occurrence of DMRs and paternal inheritance of disorders.

A multi-tissue multi-omics analysis reveals distinct kineztics in entrainment of diurnal transcriptomes by inverted feeding

Haoran Xin et al.

Summary: This study investigated the impact of inverted feeding on circadian rhythms of metabolism and physiology in peripheral tissues using global profiling of transcripts and metabolites in mice. The results showed that different tissues exhibit distinct kinetics in phase entrainment of diurnal transcriptomes by inverted feeding, with fat tissue being near-completely entrained. Despite the slow kinetics of the heart clock, fatty acid oxidation in the heart fully entrained to inverted feeding, suggesting complex mechanisms underlying circadian regulation in peripheral tissues.

The liver-clock coordinates rhythmicity of peripheral tissues in response to feeding

Gal Manella et al.

Summary: The study reveals that the hierarchy between the central and peripheral clocks in mammals has been established, and explores the disparities in responses of peripheral clocks using altered feeding paradigms and liver-clock mutant mice. Despite its significant role in nutrient processing, disruption of the liver-clock does not affect the rhythmicity of other peripheral tissues.
