The Role of miRNAs in Metabolic Diseases

Summary: Metabolic diseases, such as obesity, diabetes, dyslipidemia, and insulin resistance, are global health problems characterized by alterations in glucose and lipid metabolism. Recent studies have highlighted the important role of micro-ribonucleic acids (miRNAs) in regulating metabolic processes. miRNAs are non-coding RNAs that interact with target gene mRNAs through base pairing, ultimately inhibiting their translation. They play a significant role in various physiological processes and their involvement in the pathogenesis of metabolic diseases is continuously expanding. This review discusses the role of miRNAs in insulin production, fat metabolism, and their potential as biomarkers for predicting diabetes and MetS. Therapeutic interventions targeting miRNAs in metabolic disorders are also presented, including antisense oligonucleotides and the CRISPR/Cas9 editing platform.

Insulin-like growth factor 1 reduces coronary atherosclerosis in pigs with familial hypercholesterolemia

Summary: Injecting insulin-like growth factor 1 (IGF-1) into a pig model of coronary atherosclerosis reduced plaque burden and promoted stable plaque features. IGF-1 decreased the relative amount of coronary atheroma, increased fibrous cap thickness, and reduced necrotic core, macrophage content, and cell apoptosis. This research supports the consideration of clinical trials for IGF-1 in treating atherosclerosis.

Crosstalk Between Macrophages and Vascular Smooth Muscle Cells in Atherosclerotic Plaque Stability

Summary: Most acute cardiovascular events are caused by plaque rupture. Plaques containing large necrotic cores and thin fibrous caps are more prone to rupture, while plaques with small necrotic cores and thick fibrous caps are more protected. Understanding the cellular crosstalk and tissue microenvironment within atherosclerotic plaques is crucial for determining plaque stability and finding novel approaches to transform rupture-prone plaques into stable lesions.

Summary: Macrophage-specific overexpression of IGF-1 led to a reduction in plaque macrophages, foam cells, and atherosclerotic burden, as well as an increase in features of stable atherosclerotic plaque. This effect was likely achieved through a decrease in CXCL12-mediated monocyte recruitment and an increase in ABCA1-dependent macrophage lipid efflux.

The effect of IGF1 is mediated through the JAK2/STAT6 pathway rather than the canonical Ras/Raf/ERK or PI3K/AKT pathway. Single cell sequencing analysis shows that IGF1 treatment downregulates pro-inflammatory genes and upregulates anti-inflammatory genes in neutrophils and macrophages after MI.

Future research may focus on maximizing cardioprotective benefits while minimizing risks by considering the activation of the PI3K/Akt and ERK1/2 pathways and using biased agonists to reduce activation of the ROCK, ASK-1, and p38MAPK pathways.
