Advances in human oxytocin measurement: challenges and proposed solutions

Benjamin A. Tabak et al.

Summary: Oxytocin, a neuropeptide known for its role in reproduction and socioemotional processes, may have therapeutic potential in treating social impairments, but the precise way to manipulate this system for clinical benefit is still unknown. The inconsistent use of standardized and validated oxytocin measurement methods, including the design and study of hormone secretion and biochemical assays, presents unresolved challenges.

Sex matters: The impact of oxytocin on healthy conditions and psychiatric disorders

Donatella Marazziti et al.

Summary: Oxytocin plays a crucial role in regulating physiological processes and emotional states, with beneficial effects mediated by the autonomic and immune systems. This study aimed to explore plasma oxytocin levels in psychiatric patients and investigate sex-related differences compared to healthy controls. The results showed that healthy women had higher oxytocin levels compared to men, but sex differences were less apparent or reversed in psychiatric patients.

Establishing the content validity of the Epworth Sleepiness Scale for Children and Adolescents in Prader-Willi syndrome

Vanessa Perez Patel et al.

Summary: This study aimed to establish the content validity of the parent/caregiver version of the Epworth Sleepiness Scale for Children and Adolescents (ESS-CHAD), a measure of daytime sleepiness, in Prader-Willi syndrome (PWS) patients. Through qualitative video interviews and thematic analysis, the study found that ESS-CHAD accurately measured relevant concepts of daytime sleepiness in PWS patients and was appropriate for evaluating the treatment efficacy of daytime sleepiness in PWS.

Distinct promoter regions of the oxytocin receptor gene are hypomethylated in Prader-Willi syndrome and in Prader-Willi syndrome associated psychosis

Hannah M. Heseding et al.

Summary: This study investigated the methylation rates of the oxytocin receptor gene in individuals with Prader-Willi syndrome (PWS) and found significantly lower methylation levels, particularly in the presence of psychosis. The study also identified E2F1 as a potential transcription factor involved in the regulation of the oxytocin system.

Exploring associations between postpartum depression and oxytocin levels in cerebrospinal fluid, plasma and saliva

Qianmin Chen et al.

Summary: This study aimed to explore the association between postpartum depression (PPD) and oxytocin (OXT) levels, as well as assess the interaction between peripheral secretion and central release of OXT. The results showed that OXT concentrations in cerebrospinal fluid (CSF), plasma, and saliva were negatively correlated with PPD and could be valuable predictors of PPD. Additionally, OXT concentrations in plasma and saliva were positively correlated with CSF OXT concentrations.

Linking oxytocin and arginine vasopressin signaling abnormalities to social behavior impairments in Prader-Willi syndrome

Summary: Prader-Willi syndrome (PWS) is a genetic neurodevelopmental disorder characterized by global hypothalamic dysfunction. Dysregulation of neuropeptide signaling pathways, particularly oxytocin (OXT) and arginine vasopressin (AVP), has been implicated in the social behavior impairments seen in individuals with PWS.

Parenting stress in families of children with Prader-Willi syndrome

Shi-Bing Wong et al.

Summary: This study evaluated parenting stress and behavioral symptoms in families of children with PWS, finding a high level of stress reported by a significant portion of families. Older patients were associated with higher stress levels. Various behavioral and somatic symptoms in PWS patients were found to contribute to increased parenting stress, emphasizing the importance of dedicated medical care for PWS patients and their families.

Behavioral features in Prader-Willi syndrome (PWS): consensus paper from the International PWS Clinical Trial Consortium

Summary: Prader-Willi syndrome (PWS) is a rare neurodevelopmental genetic disorder characterized by severe hyperphagia and other behavioral challenges, impacting daily functioning and quality of life for individuals with PWS and their families. Effective therapies for these behaviors are still elusive, with a lack of consensus on how to define and measure the complex behavioral features of PWS being a limiting factor for treatment studies.

The multiple faces of the oxytocin and vasopressin systems in the brain

Summary: Traditionally, hypothalamic neuroendocrine cells were classified into magnocellular and parvocellular neurons for hormone release and brain circuit regulation purposes respectively. However, recent studies show these classic projections are outdated, with oxytocin and vasopressin released in the brain from dendrites and distant axon varicosities to modulate information transfer through conventional synapses.

Patients with PWS and related syndromes display differentially methylated regions involved in neurodevelopmental and nutritional trajectory

Juliette Salles et al.

Summary: The study focused on investigating epigenetic modifications in patients with Prader-Willi syndrome (PWS) and PWS-related disorders caused by inactivation of two genes, SNORD116 and MAGEL2, in the PWS chromosomal region. Results showed that PWS patients had differential DNA methylation patterns associated with neurodevelopmental pathways, endocrine dysfunction, and social and addictive processes, while mutations in SNORD116 and MAGEL2 were linked to metabolic pathways and nervous system development, and macromolecule biosynthesis, respectively. Epigenetic modifications varied between PWS and related disorders, suggesting relevance to different associated phenotypes.
