CRISPR interference and activation of the microRNA-3662-HBP1 axis control progression of triple-negative breast cancer

Summary: MiR-3662 is minimally expressed in normal tissues but highly expressed in various cancers, especially in TNBC and African American breast cancer. Inhibition or knockout of miR-3662 suppresses proliferation and migration of TNBC cells by reducing Wnt/β-catenin signaling activation. HMG-box transcription factor 1 (HBP-1) is identified as a target of miR-3662 responsible for TNBC cell proliferation.

CRISPRi for specific inhibition of miRNA clusters and miRNAs with high sequence homology

Summary: miRNAs are noncoding RNAs involved in gene expression regulation, and their inhibition using CRISPR/dCas9 can effectively suppress miRNA and miRNA clusters. To address the challenges in CRISPRi experiments for miRNA repression, we propose a strategy that includes prediction of miRNA transcription start sites (TSSs) and the design of sgRNAs. Compared to traditional siRNA/shRNA-based methods, CRISPRi improves the specificity of miRNA repression and enhances the consistency of silencing effects on entire miRNA clusters. This strategy can also be adapted for the activation of miRNA expression.

miR-200 deficiency promotes lung cancer metastasis by activating Notch signaling in cancer-associated fibroblasts

Summary: miR-200 miRNAs have been identified as potent suppressors of lung adenocarcinoma metastasis, with their expression specifically repressed in metastatic lung adenocarcinomas and their decrease strongly correlating with poor patient survival. miR-200 deficiency promotes the proliferation and activation of adjacent cancer-associated fibroblasts, which in turn increases the metastatic potential of lung cancer cells. The functional interaction between cancer cells and CAFs, regulated by miR-200 targeting Notch ligands, constitutes an essential mechanism to promote metastatic potential.
