Genomic Sequencing for Bladder Urothelial Carcinoma and Its Clinical Implications for Immunotherapy

Ryul Kim et al.

Summary: This study explores the genomic and transcriptomic characteristics of bladder cancer (BC) and investigates the impact of immune checkpoint inhibitors (ICI) on its treatment. The results show that a subset of patients express PD-L1 and have a higher tumor mutational burden. Furthermore, immune-responsive pathways and the CTLA4 pathway are enriched in PD-L1 positive BC. The study provides a theoretical basis for the application of dual checkpoint inhibition in bladder cancer treatment.

Copper induces cell death by targeting lipoylated TCA cycle proteins

Peter Tsvetkov et al.

Summary: Copper is an essential cofactor for all organisms, but excessive copper can lead to cell death. We found that copper-dependent cell death mechanism is different from known mechanisms, and it is related to mitochondrial respiration. Copper directly binds to lipoylated components of the tricarboxylic acid cycle, causing protein aggregation and loss of iron-sulfur cluster proteins, ultimately leading to cell death.

A Novel Cuproptosis-Related Prognostic Gene Signature and Validation of Differential Expression in Clear Cell Renal Cell Carcinoma

Zilong Bian et al.

Summary: This study systematically evaluated the genetic alterations of cuproptosis-related genes in clear cell renal cell carcinoma (ccRCC) and constructed a prognostic signature of these genes. The results suggest that the cuproptosis-related gene signature may serve as a potential prognostic predictor for ccRCC patients and is associated with immune infiltration levels and PD-1 expression.

Selective Targeting of Cancer Cells by Copper Ionophores: An Overview

Valentina Oliveri

Summary: Conventional cancer therapies face challenges due to off-target effects, leading to the need for new therapeutic agents with increased selectivity, reduced side effects, and the ability to overcome cancer cell resistance targeting cancer stem cells. Some copper ionophores show promise in inducing cuproptosis in cancer cells with greater selectivity.

Long non-coding RNA/epithelial-mesenchymal transition axis in human cancers: Tumorigenesis, chemoresistance, and radioresistance

Mehrdad Hashemi et al.

Summary: Epithelial-to-mesenchymal transition (EMT) is an important event associated with embryogenesis, tumor progression, metastasis, and therapeutic resistance. This review comprehensively discusses the role and mechanism of action of long non-coding RNAs (lncRNAs) in regulating the EMT process in various cancers, as well as their involvement in the regulation of transcription factors and therapeutic response. The review also emphasizes the role of exosomal lncRNAs in the regulation of EMT, metastasis, and therapeutic response in cancers.

Overcoming the compensatory elevation of NRF2 renders hepatocellular carcinoma cells more vulnerable to disulfiram/copper-induced ferroptosis

Xueying Ren et al.

Summary: DSF/Cu selectively exerts efficient cytotoxic effect on HCC cells, inhibits migration, invasion, and angiogenesis of HCC cells, impairs mitochondrial homeostasis, increases free iron pool, enhances lipid peroxidation, and eventually results in ferroptotic cell death. Inhibition of NRF2 elevation strengthens HCC cells more vulnerable to DSF/Cu induced ferroptosis.
