A novel microdeletion upstream of HOXD13 in a Chinese family with synpolydactyly

Weimin Jia et al.

Summary: This study investigated a Chinese family with Synpolydactyly (SPD) and found that all affected individuals in the family carry a heterozygous 11,451 bp microdeletion variant, which co-segregated with SPD in the family, suggesting it as the disease-causing variant for SPD in this family. The identified microdeletion upstream of the HOXD13 gene is the smallest deletion associated with SPD1, without altering the sequence of the HOXD13 gene.

A Novel Missense Variant of HOXD13 Caused Atypical Synpolydactyly by Impairing the Downstream Gene Expression and Literature Review for Genotype-Phenotype Correlations

Ruiji Guo et al.

Summary: Synpolydactyly (SPD) is a hereditary congenital limb malformation caused by mutations in the HOXD13 gene. Research has shown that different mutations in HOXD13 are associated with the severity and symptoms of SPD, providing insights for clinical diagnosis and genetic counseling.

Deciphering the mutational signature of congenital limb malformations

Liying Sun et al.

Summary: This study aimed to decipher the mutational signature of congenital limb malformations (CLM) using exome sequencing, identifying novel pathogenic variants and candidate genes. By expanding the phenotypic spectrum of CLM, the researchers discovered RPL9 and UBA2 as potential new genes for CLM. The study significantly increased diagnosis rates among patients with a family history of CLM or multiple affected limbs, providing valuable insights into the genetic basis of these syndromes.

An update on the neurological short tandem repeat expansion disorders and the emergence of long-read sequencing diagnostics

Sanjog R. Chintalaphani et al.

Summary: Short tandem repeat (STR) expansion disorders play a significant role in various human neurological diseases. The current laboratory techniques for diagnosing repeat expansions are not ideal, but long-read sequencing platforms show promise in improving the detection and understanding of these disorders.
