Cancer statistics, 2022

Rebecca L. Siegel et al.

Summary: Each year, the American Cancer Society compiles data on cancer occurrence and outcomes in the United States. The latest data shows that breast and prostate cancer progress has stagnated, while lung cancer has shown improvements in survival rates. Lung cancer incidence for advanced disease has declined while localized-stage rates have increased, resulting in higher survival rates. Mortality patterns align with incidence trends, with lung cancer deaths declining rapidly, breast cancer deaths slowing, and prostate cancer deaths stabilizing.

Association of genetic variants in autophagy-lysosome pathway genes with susceptibility and survival to prostate cancer

Qiuyuan Zhu et al.

Summary: This study found that the rs7573065 in the SLC11A1 gene was associated with an increased risk and poor overall survival of prostate cancer, suggesting that genetic variants in autophagy-lysosome pathway genes may serve as risk and prognostic biomarkers for prostate cancer.

Polygenic Risk Scores Associated with Tumor Immune Infiltration in Common Cancers

Jungyoon Choi et al.

Summary: Polygenic risk scores (PRSs) have been used to predict cancer risk, but their association with immune infiltration in solid tumors is still unclear. In this study, PRSs for common cancers were constructed using risk variants from previous genome-wide association studies, and their associations with immune traits were evaluated. Significant associations were identified between PRSs and immune traits, including some that were not detected in the overall analysis when stratified by stage. This study provides insights into the genetic susceptibility underlying immune response in cancer development and suggests potential applications for early diagnosis and therapeutic targeting strategies.

Downregulation of SEPTIN5 inhibits prostate cancer progression by increasing CD8+T cell infiltration

Ming Chang et al.

Summary: This study found that SEPT5 is overexpressed in prostate cancer and is associated with prognosis. Downregulation of SEPT5 inhibits the progression of prostate cancer and increases immune cell infiltration, especially CD8+ T cells. This may be achieved by promoting the production of chemokines and cytokines associated with immune cell infiltration. SEPT5 may serve as a prognostic biomarker for prostate cancer and a potential target for enhancing the efficacy of immunotherapy.

Summary: Every year, the American Cancer Society projects the numbers of new cancer cases and deaths in the United States, with the latest data showing a significant decline in lung cancer mortality, while prostate cancer mortality has plateaued and breast and colorectal cancer mortality have slowed. Improvements in treatment have accelerated progress against lung cancer, leading to a record drop in overall cancer mortality.

Role of lysosomes in physiological activities, diseases, and therapy

Ziqi Zhang et al.

Summary: Lysosomes, traditionally known as digestive organelles, have now been recognized for their diverse roles in degradation, nutrient sensing, and immunity. Numerous studies have shown the involvement of lysosome-related mechanisms in various diseases such as atherosclerosis, neurodegenerative diseases, autoimmune disorders, pancreatitis, lysosomal storage disorders, and malignant tumors. Dysregulation of lysosomal function in these diseases leads to pathological changes and affects disease progression.

Identification of a five gene signature to predict time to biochemical recurrence after radical prostatectomy

John P. Corradi et al.

Summary: Identification of novel biomarkers associated with high-risk prostate cancer or biochemical recurrence can improve detection, prognosis, and treatment. A predictive model of biochemical recurrence following radical prostatectomy was developed using a large sample of prostate specimens and validated in two external data sets.

IFI30 expression predicts patient prognosis in breast cancer and dictates breast cancer cells proliferation via regulating autophagy

Yan Fan et al.

Summary: IFI30 is highly expressed in breast cancer tissues and is associated with poor patient outcomes. Knockdown of IFI30 inhibits proliferation, migration, and invasion of breast cancer cells and significantly inhibits tumor growth in vivo. Increased accumulation of LC3-II and p62 suggests impaired autophagy in IFI30 knockdown cells.
