A Multilayered Mesoporous Gold Nanoarchitecture for Ultraeffective Near-Infrared Light-Controlled Chemo/Photothermal Therapy for Cancer Guided by SERS Imaging

Summary: A multifunctional theragnostic nanoplatform, consisting of mesoporous silica-coated gold nanostar with a cyclic Arg-Gly-Asp (RGD)-coated gold nanocluster shell, is reported. This nanoplatform exhibits enhanced SERS signals, improved NIR-induced photothermal conversion efficiency, high drug loading efficiency, and effective NIR-triggered drug release. It shows excellent performance in tumor targeting and achieves ultraeffective therapy in tumor cells and mouse models with NIR irradiation. This study offers a promising approach to effective noninvasive tumor treatment using SERS imaging and on-site triggered chemo-PTT.

Targeting integrin pathways: mechanisms and advances in therapy

Summary: Integrins are cell-adhesion receptors that play important roles in cell signaling and communication. Targeting integrins for therapeutic purposes has been challenging due to their complexity and opposing characteristics. Currently, there are only seven approved drugs targeting integrins on the market, but there are ongoing efforts to develop new therapies and diagnostic tools. Understanding the regulatory mechanisms of integrins and addressing unmet clinical needs are crucial for successful development of integrin-targeted therapies.

On-Cell Catalytic Detection of Epithelial-to-Mesenchymal Transition by a Clusterzyme Bioprobe

Summary: We have developed a peptide-conjugated metal cluster probe for quantitative analysis of a membrane protein biomarker and detection of epithelial-to-mesenchymal transition in tumor cells. This probe, called clusterzyme, has selective recognition and enzyme-like activity, allowing sensitive analysis of the membrane protein on cells. This method is compatible with traditional enzyme-linked immunosorbent assays and offers improved detection sensitivity and accuracy. The expression level of the membrane protein reflects the migration and invasion ability of tumor cells, revealing the epithelial-to-mesenchymal transition process. This work provides a simple and sensitive approach to monitor tumor cell type evolution at the molecular level, with potential applications in early cancer diagnosis and therapy assessment.

Summary: CuCs is a novel antibacterial molecule with broad-spectrum and excellent antibacterial activity, able to disrupt bacterial wall structure, regulate GSH/GSSG ratio, induce reactive oxygen species production, and ultimately lead to bacterial death. In vivo studies in mice demonstrate that CuCs can significantly treat skin wound infections and sepsis caused by MRSA, with similar therapeutic efficacy as mupirocin ointment and vancomycin. Compared to silver and platinum clusters, CuCs show extremely low cytotoxicity to normal mammalian cells.
