Mitochondrial regulation in spermatogenesis

Summary: The classic roles of mitochondria in energy production, metabolism, and apoptosis are well known. However, recent evidence suggests that mitochondria also play a role in regulating stem cell fate decision and lineage commitment through signaling transduction, protein modification, and epigenetic changes. This is particularly important in spermatogenesis, where germ cells have changing metabolic needs during development. It is now recognized that proper male fertility relies on controlled plasticity of mitochondrial features and functions. The unique role of mitochondria in germ cell ncRNA processing adds another layer of complexity to mitochondrial regulation during spermatogenesis. In this review, we discuss potential regulatory mechanisms of how mitochondria reshape their features, activities, and functions to support germ cell fate transitions during spermatogenesis. We also review recent findings on the coordination between mitochondrial regulators and germline proteins in germ cell-specific activities.

COX4-like, a Nuclear-Encoded Mitochondrial Gene Duplicate, Is Essential for Male Fertility in Drosophila melanogaster

Knockdown and knockout of COX4L lead to male sterility in Drosophila, indicating its essential role in sperm development. Furthermore, COX4L is involved in the embryonic development and viability of Drosophila, highlighting its significance in overall reproductive success.

Single-cell RNA-sequencing reveals pre-meiotic X-chromosome dosage compensation in Drosophila testis

The Drosophila orthologue of the primary ciliary dyskinesia-associated gene, DNAAF3, is required for axonemal dynein assembly

Summary: Ciliary motility in humans relies on highly conserved axoneme-specific dynein motor complexes, and mutations in these motors can lead to primary ciliary dyskinesia (PCD). Research in Drosophila has identified a less-known assembly factor, DNAAF3, which is essential for the function of mechanosensory neurons and sperm-producing cells in fruit flies. Mutations in DNAAF3 result in deaf larvae and uncoordinated adults, indicating a dysfunction in Ch neurons. Additionally, the absence of dynein arms in mutant Ch neuron cilia and immotile sperm with disrupted axoneme further demonstrates the importance of dynein motors in ciliary function and sperm motility.
